肛门失禁动物模型基因转录组学分析

摘  要:目的通过转录组学测序技术(RNA-seq)分析手术离断括约肌致猪肛门失禁动物模型肛门括约肌的转录组学变化。方法采用随机数字表法将8只成年长白猪分为空白组、肛门失禁动物模型组(模型组),每组4只。模型组予以手术离断括约肌致肛门括约肌损伤方式构建肛门失禁模型,空白组未手术。取各组肛门括约肌组织,提取肛门括约肌组织总RNA,采用RNA-seq技术进行转录组学测序,进行GO(Gene oncology)和KEGG(Kyoto Encyclopedia of Genes and Genomes)生物信息学分析,并筛选差异基因。结果测序结果表明,模型组与空白组差异基因共431个,其中上调基因280个,下调基因151个。转录组学测序分析发现模型组中蒙脱石基因(OBSCN)、趋化因子配体(CCL26)、Fyn-结合蛋白1(FYB1)及CXC基序趋化因子10(CXCL10)等基因表达上调,α-肌动蛋白-2(ACTG2)、人平滑肌肌球蛋白轻链激(MYLK)、鸡鸭甲状腺激素应答基因(THRSP)、活性物质2(ACTA2)等基因表达下调。GO分析发现,上调表达基因主要与免疫反应和趋化性等基因功能相关。KEGG分析发现,上调表达基因主要属于金黄色葡萄球菌感染、抗原处理和提呈相关基因等信号通路。结论转录组学分析提示OBSCN等上调基因可能参与肛门失禁发生发展过程,这为研究肛门失禁致病机制研究提供实验依据。Objective To analyze the transcriptome changes of anal sphincter in an animal model of porcine anal incontinence by RNA-Seq.Methods 8 landwhite pigs were randomly by random number table divided into blank group and anal incontinence animal model group,with 4 pigs in each group.The anal incontinence model was established in the model group by surgical amputation of sphincter and anal sphincter injury,while the control group was not operated.Total RNA was extracted from anal sphincter tissue of the each group,and transcriptome sequencing was performed with using RNA-Seq technology.Gene oncology(GO)and kyoto encyclopedia of genes and Genomes(KEGG)bioinformatics analysis were performed,and the different genes were screened and validated.Results There were 431 differentially expressed genes inmodel group andblank group,including 280 up-regulated genes and 151 down-regulated genes.Transcriptome sequencing analysis showed that OBSCN gene,CCL26 gene,FYB1 gene and CXCL10 gene were up-regulated,α-actin 2(ACTG2),human smooth muscle myosin light chain stimulation(MYLK),thyroid hormone responsive gene of chicken and duck(THRSP),ACTA2 and other genes were down-reg-ulatedin animal model group of anal incontinence.GO analysis showed that the up-regulated genes were mainly relat-ed to the function of immune response and chemotaxis.KEGG analysis showed that the up-regulated genes mainly be-longed to the signaling pathways of ST aphylococcus aureus infection,antigen processing and presentation relatedgenes.ConclusionTranscriptomic analysis suggests that up-regulated genes such as OBSCN may be involved in thedevelopment of incontinence,which provides an experimental basis for studying pathogenesis of incontinence.
