金银花与灰毡毛忍冬治疗流感病毒FM1肺炎小鼠作用比较

摘　　要：目的:研究金银花和灰毡毛忍冬对流感病毒FM1所致肺炎小鼠的治疗作用和抗流感病毒功效差异。方法:建模给药后第2、第4、第6天分别测定金银花和灰毡毛忍冬水提液对肺炎小鼠的肺部炎症抑制作用、死亡保护作用、肺组织的病毒载量、病理切片变化、炎症因子肿瘤坏死因子α(TNF-α)和白细胞介素(IL-6)基因表达的变化,动态、系统地评价二者抗病毒功效。结果:二者都可抑制肺炎小鼠体质量的下降,对小鼠的死亡保护率分别为31.43%和12.61%,第6天肺指数抑制率分别为30.89%和17.89%,都能抑制肺组织病毒载量,逆转TNF-α和IL-6种炎症因子mRNA升高,小鼠肺组织病变明显改善,偶见炎症细胞浸润,灶性分布的病理改变消失,且金银花治疗效果略优于灰毡毛忍冬。结论:金银花和灰毡毛忍冬都能抑制流感病毒FM1感染造成的小鼠肺损伤,但金银花治疗效果略优于灰毡毛忍冬。Objective:To study the therapeutic effect of Lonicera japonica Flos and Lonicera macranthoides Hand.-Mazz.on pneumonia mice caused by influenza virus FM1 and the difference of anti-influenza efficacy.Methods:On the 2nd day,4th day and 6th day after model administration,the inhibitory effect of Lonicera japonica Flos and Lonicera macranthoides Hand.-Mazz.water extracts on lung inflammation,the protective effect against death,the viral load of lung tissue,the changes of pathological sections and the changes of inflammatory factors,tumor necrosis factorα(TNF-α)and interleukin-6(IL-6)gene expression were measured,and the antiviral effects of them were dynamically and systematically evaluated.Results:Both of them could inhibit the weight loss of pneumonia mice,with the death protection rates of 31.43%and 12.61%respectively,and the lung index inhibition rates of 30.89%and 17.89%on the 6th day,respectively.Both of them could inhibit the viral load of lung tissue,reverse the increase of TNF-αand IL-6 inflammatory factors mRNA,and obviously improve the lung tissue pathological changes of mice,with occasional infiltration of inflammatory cells,disappearance of pathological changes of focal distribution.And the therapeutic effect of Lonicera japonica Flos is slightly better than that of Lonicera macranthoides Hand.-Mazz..Conclusion:Both of Lonicera japonica Flos and Lonicera macranthoides Hand.-Mazz.can inhibit the lung injury caused by influenza virus FM1 infection in mice,but the therapeutic effect of Lonicera japonica Flos is slightly better than that of Lonicera macranthoides Hand.-Mazz..
