摘 要:目的:为了探讨不同浸泡条件对杜仲雄花水提液中各活性成分的影响,并评价其对NO_(2)^(-)的清除效果。方法:采用分光光度法、高效液相色谱法分别测定杜仲雄花水提液总黄酮、绿原酸、京尼平苷酸、桃叶珊瑚苷4种活性成分的含量变化,并使用酶标仪进一步测定NO_(2)^(-)清除率。结果:不同浸泡时间对4种活性成分溶出量的影响均存在显著性差异(P<0.05),直接提取效果优于其他浸泡方式,综合考虑在45~60min时各功效成分含量最高,当浸泡温度保持在90 ℃以上时更有利于活性成分的溶出。将杜仲雄花水提液用于NO_(2)^(-)清除,结果显示直接提取清除效果最佳,清除率达64.4%±3.23%。90℃和95℃对NO_(2)^(-)清除率影响差异性不显著(P>0.05),但均比85℃清除效果好,在提取60min时清除率最高为68.02%±4.33%。结论:不同浸泡方式对杜仲雄花有效成分的溶出量影响较显著,在温度90℃以上,直接浸提45~60min可以最大限度的获取其功能功效成分,并且对NO_(2)^(-)具有较强的清除作用。Objective:To investigate the effects of different soaking conditions on the active components in the aqueous extract of Eucommia ulmoides male flowers,and to evaluate its scavenging effect on NO_(2)^(-).Methods:the contents of total flavonoids,chlorogenic acid,geniposide and aucubin in the aqueous extract of Eucommia ulmoides male flower were determined by spectrophotometry and HPLC,and the clearance of NO_(2)^(-) was further determined by enzyme labeling instrument.Results:there were significant differences in the effects of different soaking time on the dissolution of four active components(P<0.05),the effect of direct extraction was better than other soaking methods.Considering that the content of each functional component was the highest within 45~60min,and it was more conducive to the dissolution of active components when the soaking temperature was maintained above 90℃.When the aqueous extract of Eucommia ulmoides male flower was used for NO_(2)^(-)removal,the results showed that the effect of direct extraction was the best,and the removal rate was 64.4%±3.23%.90℃ and 95℃ had a poor effect on NO_(2)^(-)removal rate,the difference was not significant(P>0.05),but the scavenging effect was better than 85℃,and the highest scavenging rate was 68.02%±4.33% when extracted for 60 min.Conclusion:different soaking methods had significant effects on the dissolution of the effective components of Eucommia ulmoides male flowers.When the temperature was above 90℃,direct extraction for 45~60min could obtain the functional components to the greatest extent,and had a strong scavenging effect on NO_(2)^(-).
