脑清通颗粒对肝热痰瘀证高血压模型大鼠血清hs-CRP及血管形态的影响

摘　　要：目的观察脑清通颗粒对肝热痰瘀高血压模型大鼠血清超敏C反应蛋白(Hypersensitive C-reactive protein,hs-CRP)及血管形态的影响。方法采用随机数字表法将60只SD大鼠随机分成空白对照组、模型对照组、西药组和中药大、中、小剂量组。正常对照组除给予生理盐水10 ml/kg灌胃外,不进行其他任何干预措施。其余各组均给予环孢菌素A 25 mg/kg皮下注射(2 ml/kg)、高脂乳剂灌胃(10 ml/kg)、附子溶液灌胃(相当于2 g生药/kg)进行造模,并灌胃给药1次/d,模型对照组给予生理盐水10 ml/kg,西药组给予卡托普利10 mg/kg,中药大、中、小剂量组分别给予脑清通颗粒5.2 g/kg、2.6 g/kg、1.3 g/kg,灌胃量均为10 ml/kg,1次/d,连续21 d。结果与空白对照组比较,模型对照组对照组血压和血清中hs-CRP升高,差异有统计学意义(P<0.01),光镜下显示模型对照组血管形态明显紊乱;与模型对照组比较,西药组、中药大剂量组、中药中剂量组、中药小剂量组均可降低血压和血清hs-CRP,血压下降以第3周为著,均具有统计学意义(P<0.05,P<0.01);西药组、中药大剂量组、中药中剂量组血管中、内膜不同程度改善,中药大剂量组改善最为明显。结论脑清通颗粒可使肝热痰瘀证高血压大鼠血清炎症反应降低,此机制可能是中药调整血压,改善高血压血管重塑的重要环节。Objective To observe the effects of Naoqingtong Granules on serum high-sensitivity C-reactive protein(hs-CRP)and vascular morphology in hypertension model rats with liver heat and phlegm stagnation syndrome.Methods Sixty SD rats were randomly divided into a normal control group,a model control group(normal saline,10 ml/kg),a western medicine(captopril,10 mg/kg)group,and high-,medium-and low-dose Chinese medicine groups(Naoqingtong Granules,5.2 g/kg,2.6 g/kg,and 1.3 g/kg)according to a random number table.The rats in the normal control group received normal saline at 10 ml/kg by gavage,while those in the remaining groups were treated with 25 ml/kg cyclosporin A by subcutaneous injection(2 ml/kg),high-fat emulsion by gavage(10 ml/kg),and Aconiti Lateralis Radix Praeparata solution by gavage(equivalent to 2 g/kg crude drug)for model establishment.Oral administration was carried out once per day.The dosage for oral administration was 10 ml/kg,once per day for 21 days.Results Compared with the blank control group,the model control group showed increased blood pressure and serum hs-CRP(P<0.01)and disorderedvascular morphologyunder the light microscope.Compared with the model control group,the groups with drug intervention showed reduced blood pressure and serum hs-CRP,and the decrease in the blood pressure was significant in the 3 rd week(P<0.05,P<0.01).The western medicine group,the high-,and medium-dose Chinese medicine groups showed improved vascular media and intima to varying degrees,especially in the high-dose Chinese medicine group.Conclusion Naoqingtong Granules can reduce the serum inflammatory response in hypertension rats with liverheat and phlegm stagnation syndrome,and the mechanism may be an important link of Chinese medicine to adjust blood pressure and improve vascular remodeling in hypertension.
