摘  要:目的证明白细胞介素24(interleukin-24,IL-24)在人血管内皮细胞(human vascular endothelial cells,HECV)的受体表达,评估IL-24对人血管内皮细胞迁移及生长的影响。方法通过细胞功能试验,检测重组人白细胞介素24(recombinant human interleukin 24,rhIL-24)对人血管内皮细胞的生长、迁移、跨内皮阻力的影响。结果IL-24和受体分子被发现在人血管内皮细胞中有表达。用rhIL-24对这一细胞株进行处理,发现能促进细胞迁移率但不影响细胞生长或测试浓度的渗透性。结论rhIL-24能促进人血管内皮细胞的迁移,但不影响细胞生长或测试浓度的渗透性。本研究证实这些影响与IL-24和蛋白激酶B(protein kinase B,AKT)或者阻断磷脂酶Cγ(blocking phospholipase C,PLCγ)相关通道之间的潜在联系有关。Objective To demonstrate the receptor expression of Interleukin-24(IL-24)in human vascular endothelial cells(HECV);to evaluate the effect of IL-24 on the migration and growth of human vascular endothelial cells.Methods The effects of recombinant human interleukin 24(rhIL-24)on the growth,migration and transendothelial resistance of human vascular endothelial cells were detected by cell function test.Results IL-24 and receptor molecules were found to be expressed in human vascular endothelial cells.Treatment of this cell line with rhIL-24 was found to promote cell mobility without affecting cell growth or permeability at the tested concentrations.Conclusion rhIL-24 can promote the migration of human vascular endothelial cells,but does not affect cell growth or permeability of tested concentrations.This study confirms that these effects are related to potential links between IL-24 and protein kinase B(AKT)or blocking phospholipase Cγ(PLCγ)related channels.
