摘  要:目的探讨依达拉奉右莰醇对大鼠液压冲击脑损伤模型的神经保护作用。方法选取雄性SD大鼠96只,随机分为假手术组、脑损伤组、依达拉奉右莰醇组,每组32只。假手术组大鼠只行麻醉后颅骨开窗,不行液压冲击处理。脑损伤组和依达拉奉右莰醇组大鼠进行液压冲击处理,并在建模成功后,依达拉奉右莰醇组大鼠用经尾静脉注射给药方法,每次剂量为3 mg/kg,于造模后12 h、24 h、3 d、7 d连续给药4次;假手术组和脑损伤组大鼠则注射等量生理盐水。在造模后12 h、24 h、3 d、7 d,采用干湿质量法测定脑组织含水量,采用酶联免疫吸附试验(ELISA)检测白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)的表达水平,用二氨基联苯胺标记并定量分析单链DNA(ssDNA)、羟基脲(Hu)、8-羟化脱氧鸟苷(8-OHdG)或四羟壬烯醛(4-HNE)阳性细胞数量,用Hu或GFAP双免疫荧光法对脑组织损伤区周围ssDNA进行双免疫荧光染色。结果与假手术组比较,脑损伤组大鼠各时间点脑组织含水量均明显增加,炎性因子IL-1β、IL-6和TNF-α水平均明显升高,差异均有统计学意义(P<0.05)。与脑损伤组比较,依达拉奉右莰醇组大鼠各时间点脑组织含水量均明显降低,炎性因子IL-1β、IL-6和TNF-α水平均明显降低,差异均有统计学意义(P<0.05)。与假手术组比较,脑损伤组各时间点脑组织中ROS含量升高、SOD含量降低,差异均有统计学意义(P<0.05);与脑损伤组比较,依达拉奉右莰醇组大鼠各时间点脑组织中ROS含量降低、SOD含量升高,差异均有统计学意义(P<0.05)。依达拉奉右莰醇组大鼠各时间点脑组织损伤区域周围8-OHdG、4-HNE、ssDNA阳性细胞数量少于脑损伤组,差异有统计学意义(P<0.05)。脑损伤后,随着时间的推移,Hu阳性细胞数量逐渐增多,在第7天时最显著,且依达拉奉右莰醇组高于脑损伤组,差异有统计学意义(P<0.05)。在脑损伤组中�Objective To investigate the neuroprotective effect of edaravone dexcamphenol against hydraulic shock brain injury in rats.Methods A total of 96 male SD rats were randomly divided into sham operation group,brain injury group and edaravone dexcamphenol group,with 32 rats in each group.The rats in sham operation group only received craniotomy after anesthesia.The rats in brain injury group and edaravone dexcamphenol group received hydraulic shock treatment,and the rats in edaravone dexcamphenol group were given 3 mg/kg edaravone dexcamphenol via tail vein at 12 h,24 h,3 d and 7 d after successful modeling.The rats in sham operation group and brain injury group were injected with the same amount of normal saline.The indexes were observed at 12 h,24 h,3 d and 7 d after modeling.The moisture content in brain tissue at each time point was measured by dry-wet mass method.The expression levels of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)were detected by enzyme-linked immunosorbent assay(ELISA)at each time point.Diaminobenzidine was used to label and quantitatively analyze the number of ssDNA,Hu,8-OHdG or 4-HNE positive cells.Hu or GFAP double immunofluorescence method was used to detect ssDNA around brain tissue injury.Results Compared with sham operation group,the water content in brain tissue and the levels of inflammatory factors IL-1β,IL-6 and TNF-αwere significantly increased at each time point in brain injury group(P<0.05).Compared with brain injury group,the water content in brain tissue in edaravone dexcamphenol group was significantly decreased at each time point,and the levels of inflammatory factors IL-1β,IL-6 and TNF-αwere significantly decreased(P<0.05).Compared with sham operation group,ROS content was increased and SOD content was decreased in the brain tissue at each time point in brain injury group(P<0.05).Compared with brain injury group,ROS content was decreased and SOD content was increased at each time point in edaravone dexcamphenol group(P<0.05).The numbers
