摘  要:目的观察脂联素(adiponectin,APN)预处理对高糖诱导下人视网膜微血管内皮细胞(HRMECs)凋亡及增殖的影响。方法将生长状况良好的HRMECs随机分为对照组(DMEM培养基,无干预)、高糖组(DMEM培养基+25 mmol/L D-葡萄糖)、高糖+APN组(细胞先用10μg/ml APN预处理2 h,然后用高糖培养基培养)。各组分别培养24 h后,Western blot检测HRMECs凋亡过程中Bax、Bcl-2以及Caspase-3的蛋白表达水平,流式检测HRMECs凋亡率,CCK-8法检测HRMECs增殖。结果与对照组比较,高糖组HRMECs的Bax、Caspase-3蛋白水平以及细胞凋亡率均明显增高,但Bcl-2的表达量及细胞增殖率均明显降低(均P<0.001)。与高糖组比较,高糖+APN组HRMECs的Bax、Caspase-3蛋白水平以及细胞凋亡率有所回落,Bcl-2的蛋白水平及细胞增殖率有所回升(均P<0.001)。结论高糖诱导下HRMECs凋亡增加,增殖减少,APN预处理可以抑制凋亡,促进增殖,提示APN对高糖下视网膜微血管内皮细胞损伤有保护作用。Objective To observe the effect of adiponectin(APN)pretreatment on apoptosis and proliferation of human retinal microvascular endothelial cells(HRMECs)under the high glucose.Methods HRMECs with good growth were randomly divided into three groups:control group,high glucose group,high glucose+APN group.Cells in control group were cultured in DMEM medium for 24 h,without intervention.Cells in high glucose group were cultured in DMEM medium and 25 mmol/L D-glucose for 24 h.Cells in high glucose+APN group were pretreated with 10μg/ml APN for 2 h and then cultured with high glucose medium for 24 h.The protein expression levels of Bax,Bcl-2,and Caspase-3 during apoptosis of HRMECs were measured by Western blot.The apoptosis rate of HRMECs was detected by flow cytometry.The proliferation of HRMECs was detected by CCK-8.Results Compared to control group,the protein levels of Bax,Caspase-3 and the apoptosis rate of HRMECs were significantly increased in high glucose group,but the expression of Bcl-2 and the proliferation rate of cells were significantly reduced(all P<0.001).Compared with high glucose group,the protein levels of Bax,Caspase-3 and the apoptosis rate of HRMECs were decreased in high glucose+APN group,while the expression of Bcl-2 and the proliferation rate of cells were increased(all P<0.001).Conclusion The apoptosis of HRMECs increases and the proliferation decreases under a high glucose condition.APN pretreatment can inhibit the apoptosis and promote the proliferation of HRMECs,suggesting that APN has protective effect against high glucose-induced retinal vascular endothelial cell damage.
