摘  要:目的探讨鼻鼽合剂对过敏性鼻炎(allergic rhinitis,AR)模型小鼠体内Th17/Treg失衡的影响。方法20只BALB/c雌性小鼠利用卵清蛋白溶液构建小鼠AR模型,分为模型组、鼻鼽合剂低剂量组、鼻鼽合剂高剂量组和辛芩组;另外5只为空白对照组。观察小鼠在实验过程中的精神状态、饮食饮水、活动情况以及毛发光泽度等情况以及对行为学症状分级进行评分。采用苏木精—伊红染色法(hematoxylin-eosin staining,HE)观察小鼠鼻黏膜组织变化。采用实时定量聚合酶链式反应(quantitative real time-polymerase chain reaction,qRT-PCR)检测小鼠肺组织中白细胞介素4(interleukin-4,IL-4)、白细胞介素17(interleukin-17,IL-17)、γ干扰素(interferon-γ,IFN-γ)、叉头样转录因子3(forkhead box p3,Foxp3)mRNA相对表达水平。结果与空白对照相比,造模后的小鼠精神萎靡,采食与饮水次数减少,出现竖毛的情况,行为学症状分级评分显示造模成功。与模型组比较,鼻鼽合剂低、高剂量组以及辛芩组的小鼠精神、饮食及毛发情况均有所改善。HE染色结果显示,模型组鼻黏膜组织可见大量炎性细胞浸润,上皮细胞排列不齐;鼻鼽合剂低、高剂量组以及辛芩组的炎性细胞明显减少。qRT-PCR结果显示,与对照组相比,模型组鼻黏膜组织中的IL-4、IL-17、IFN-γ和Foxp3的mRNA表达水平显著升高(P<0.05);与模型组比较,鼻鼽合剂低、高剂量组以及辛芩组的IL-4和IL-17的mRNA表达水平显著降低(P<0.05),但IFN-γ和Foxp3的mRNA表达水平显著上升(P<0.05);鼻鼽合剂的低剂量组和高剂量组IL-4和IL-17的mRNA表达水平则都显著低于辛芩组(P<0.05),而IFN-γ和Foxp3的mRNA表达水平则都显著高于辛芩组(P<0.05)。另外,鼻鼽合剂高剂量组的IL-4和IL-17的mRNA表达量显著低于低剂量组(P<0.05),而IFN-γ和Foxp3的mRNA表达水平都显著高于低剂量组(P<0.05)。结论鼻鼽合剂能通过调节体内Th17/Treg的失衡进而缓解小�Objective To investigate the effect of Biqiu mixture on Th17/Treg imbalance in allergic rhinitis(AR)model mice.Methods Twenty BALB/c female mice were treated with ovalbumin solution to establish a mouse AR model and divided into model group,low-dose group,high-dose group and Xinqin group,and another 5 mice were chosen as control group in addition.The mental state,diet,drinking water,activity,hair gloss of mice and the behavioral symptoms were observed and calculated during the experiment.Hematoxylin-eosin staining(HE)was used to observe the change of the nasal mucosal tissues,and quantitative real time-polymerase chain reaction(qRT-PCR)was used to detect the mRNA expression of interleukin-4(IL-4),interleukin-17(IL-17),interferon-γ(IFN-γ)and Forkhead box p3(Foxp3)in all groups.Results Compared with control group,the mice were depressed after modeling,the times of eating and drinking water were reduced,and the bristling occurred.The grading score of behavioral symptom showed that the modeling was successful.Compared with model group,the spirit,diet and hair condition of mice were improved in low and high dose groups and Xinqin group.HE staining showed that a large number of inflammatory cells infiltrated in the nasal mucosa in model group,and the epithelial cells were arranged irregularly.The inflammatory cells were decreased significantly in low,medium and high dose groups and Xinqin group.Compared with control group,IL-4,IL-17,IFN-γ,and Foxp3 in nasal mucosa were increased significantly in model group(P<0.05).Compared with model group,the expression levels of IL-4 and IL-17 were significantly reduced in low and high dose groups and Xinqin group(P<0.05),but the expression levels of IFN-γand Foxp3 were significantly elevated(P<0.05).IL-4 and IL-17 were significantly lower in low-dose and high-dose groups than those in Xinqin group(P<0.05),while IFN-γand Foxp3 were significantly higher(P<0.05).In addition,IL-4 and IL-17 in high-dose group were significantly lower in high-dose group than in low-dose group(P<0.05
