摘 要:目的天然蚕丝降解缓慢,本实验采用再生蚕丝加以调节。探讨通过天然蚕丝混合再生蚕丝纬编针织网支架覆盖骨髓间充质干细胞(BMSCs)细胞片,观察BMSCs在支架上的生长情况,并检测支架的机械强度。方法提取兔BMSCs,通过添加维生素C诱导制备BMSCs细胞片。天然蚕丝混合再生蚕丝通过纬编针织法制备网状支架,并将细胞片覆盖于支架上成支架-细胞片复合物,体外培养2周,观察细胞生长情况,并测试支架-细胞片复合物的机械强度。结果提取的BMSCs生长旺盛。黏附于支架的BMSCs呈立体生长,增殖良好。支架-细胞片复合物培养2周后的最大载荷、拉伸强度、弹性模量分别为(118.41±13.23)N、(42.78±4.63)MPa、(191.31±19.92)MPa,机械强度良好。结论天然蚕丝混合再生蚕丝针织网-细胞片复合物具有良好的细胞相容性与机械强度,有望作为组织工程韧带的支架材料。【Objective】Natural silk degrades slowly,so the regenerated silk was used to adjust its degradation rate.Bone marrow-derived mesenchymal stem cell(BMSCs)sheet were covered on mixed natural and regenerated silk weft-knitted mesh scaffold.Then the growth of BMSCs on the scaffold was observed,and the mechanical strength of the scaffold was detected.【Methods】Rabbit BMSCs were extracted,and BMSCs sheet was prepared by adding vitamin C.At the same time,the meshed scaffold was prepared by weft-knitting method of natural silk mixed regenerated silk.Then the BMSCs sheet was covered on the meshed scaffold to form scaffold-cell sheet composite.After two weeks of cultivation,the cell growth was observed,and the mechanical properties of the composite were tested.【Results】The extracted BMSCs grew well,and the BMSCs adhered to the scaffolds showed stereoscopic growth and good proliferation.The maximum load,tensile strength,and elastic modulus of the scaffold-BMSCs sheet composite cultured for two weeks were(118.41±13.23)N,(42.78±4.63)MPa,and(191.31±19.92)MPa,respectively.【Conclusion】Natural silk mixed regenerated silk mesh-BMSCs sheet composite has good cytocompatibility and mechanical strength,which is expected to be used as scaffold material for tissue engineered ligament.
