中药益精方对精索静脉曲张大鼠精子质量的影响

摘　　要：目的 观察中药益精方对精索静脉曲张大鼠精子质量的影响。方法 雄性SD大鼠30只,随机分为假手术对照组(假手术组) 10只、精索静脉曲张模型组(模型组) 10只和益精方治疗组(治疗组)10只。治疗组给予中药益精方灌胃,每日1次,共8周。对大鼠进行精液分析,并通过HE染色、TUNEL染色检测、ELISA等方法,研究精曲小管内生精细胞情况及丙二醛(MDA)、精子DNA碎片率等指标。结果 益精方颗粒治疗组与模型组比较,精索静脉曲张大鼠睾丸约翰逊评分(Johnson score)(9.2±0.51 vs 8.1±0.42 P<0.05)、精子活率(59.81±5.27 vs 53.32±4.95 P<0.05)、附睾MDA含量低(7.53±1.86 vs 10.40±2.58 mmol/g P<0.05)、精子DNA碎片率(13.30±2.39 vs 18.36±3.29%P<0.01)、睾丸生精细胞凋亡率(2.87±1.21 vs 5.2±1.74%P<0.05)均有显著差异。结论 中益精方治疗能提高精索静脉曲张大鼠的精子质量,其抗氧化作用可能在精索静脉曲张大鼠的睾丸保护中扮演重要的角色。Objective To evaluate the effects of Chinese medicine Yijing Fang on sperm quality in varicocele rats.Methods Thirty male SD rats were randomly divided into three groups including sham operation control group(n=10),varicocele model group(n=10) and Yijing Fang treatment group(n=10). The rats in the treatment group was given with Yijing Fang once a day for 8 weeks. Semen quality of rats were analyzed, and spermatogenic cells, sperm DNA fragmentation rate, malondialdehyde(MDA), and other indicators were studied by HE staining, TUNEL staining and ELISA.Results Compared with that in the model group, the Johnson score of testicular varicocele rats in Yijing Fang treatment group was increased(9.2±0.51 vs 8.1±0.42 P<0.05);Sperm viability was also increased(59.81±5.27 vs 53.32±4.95 P<0.05),whereas the content of MDA in epididymis(7.53 ± 1.86 vs 10.40 ± 2.58 mmol/g P<0.05), sperm DNA fragmentation rate(13.30±2.39 vs 18.36±3.29 % P<0.01), the apoptosis rate of spermatogenic cells were all decreased(2.87±1.21 vs 5.2±1.74 %P< 0.05). Conclusion Chinese medicine Yijing Fang can improve the sperm quality of varicocele rats, and its anti-oxidative effect may play an important role in testicular protection of varicocele rats.
