摘 要:常规的心肌组织芯片(HOC)培养的心肌细胞取向杂乱、成熟度低、机能复现程度不足,导致实验结果无法反映药物对原生心肌组织的作用。针对该问题,设计并制备了含片上机械激励的心肌组织芯片,其由拉伸薄膜、聚己内酯(PCL)支架与基底组成,通过Comsol仿真确定结构关键尺寸,使用脱模技术制造含片上机械激励的心肌组织芯片。芯片可通过循环拉伸组织诱导细胞取向、成熟。开展了阵列化细胞培养实验,验证了纤维支架上的人源心肌细胞在机械激励(拉伸应变分别为10%和20%、频率为1 Hz、连续168 h)的环境下,促进了肌节取向,并提升了Cx-43与cTnT的基因表达水平。Cardiomyocytes cultured by conventional heart-on-a-chip(HOC)have disarray orientation,low maturity and insufficient functional reappearance,leading to the failure of experimental results to reflect the effect of drugs on primary myocardial tissue.To solve the problem,a HOC with on-chip mechanical stimulator was designed and prepared.It is composed of stretched thin film,polycaprolactone(PCL)scaffold and substrate.The key dimensions of the structure were determined by Comsol simulation.The HOC with mechanical stimulator was fabricated by demoulding technique.The chip can induce cell orientation and maturation by circulating tissue stretching.Array cell culture experiments were carried out.It is verified that human cardiomyocytes on fiber scaffold promotes sarcomere orientation and enhances gene expression levels of CX-43 and cTnT under mechanical stimulation(10%and 20%tensile strains at 1 Hz frequency for 168 h).
