摘 要:为进一步探究兰坪乌骨绵羊乌质性状形成机理,该试验选取兰坪乌骨绵羊和来自兰坪的普通绵羊各50只作为样本,基于前期全基因组测序筛选的SLC17A8基因(solute carrier family 17 member 8)在兰坪乌骨绵羊具有12个高度连锁、高等位基因频率的SNP,故选择该基因作为候选基因,设计11对特异性引物对2个绵羊类群SLC17A8基因12个外显子及其侧翼区进行PCR扩增;通过紫外可见光谱法测定血液黑色素指标(TYR活力、血浆比色、脱黑色素、碱溶性黑色素、真黑色素/总黑色素);分析SLC17A8基因多态性与兰坪乌骨绵羊乌质性状的关联性。结果显示:除脱黑色素外,兰坪乌骨绵羊与普通绵羊其他各项黑色素指标差异显著(P<0.05),兰坪乌骨绵羊TYR活力、血浆比色、真黑色素/总黑色素显著高于普通绵羊;关联分析表明,EX1-G139A位点上CC型绵羊、EX2-C87A位点上AA型绵羊、EX4-C149T位点和EX4-T158C位点上CT型绵羊及EX7-C181T位点和EX7-T195C位点上TT型绵羊TYR活力均显著高于其它基因型绵羊(P<0.05),综上所述,CC、AA、CT和TT型为兰坪乌骨绵羊的优势基因型。To further explore the formation mechanism of the black-bone characteristics of the Lanping Black-bone sheep,in this experiment,50 Lanping Black-bone sheep and 50 ordinary sheep from Lanping were selected as samples.Because the SLC17A8 gene(solute carrier family 17 member 8)screened by previous whole-genome sequencing in Lanping Black-bone Sheep had 12 highly linked SNPs with high gene frequency,this gene was selected as a candidate gene.Eleven pairs of specific primers were designed for PCR amplification of 12 exons and their flank regions of SLC17A8 gene in two sheep populations.Blood melanin indexes(TYR activity,plasma colorimetry,demelanin,alkali-soluble melanin,eumelanin/total melanin)were determined by UV-Vis spectroscopy.The relationship between SLC17A8 gene polymorphism and melanin characteristics were analyzed.The results showed except for demelanin,there were significant differences in other melanin indexes between Lanping Black-bone Sheep and common sheep(P<0.05),and the TYR vitality,plasma colorimetry and ratio of true melanin to total melanin of Lanping Black-bone sheep were significantly higher than those of ordinary sheep.Correlation analysis results revealed that the TYR activity of CC sheep on site EX1-G139A,AA sheep on site EX2-C87A,CT sheep on site EX4-C149T and EX4-T158C,and TT sheep on site EX7-C181T and EX7-T195C was significantly higher than that of other genotype(P<0.05).In conclusion,CC,AA,CT and TT types are the dominant genotype of Lanping Black-bone sheep.
