摘 要:藏山羊是中国独特的地方品种。为进一步从基因组水平上了解藏山羊的遗传多样性及群体遗传结构,该研究对西藏昌都黑山羊的3个不同地方群体进行了全基因组重测序。鉴定3个采样群体的SNP位点结果表明,共鉴定出13426331个SNP位点;遗传多样性及连锁不平衡分析结果显示,3个采样群体的核苷酸多样性相近,并且LD衰减曲线相似;通过NJ进化树分析,其拓扑结构显示3个采样群体聚成1支,与西藏地区其它8个山羊品种或群体显著分离;群体结构分析结果显示,采样群体间亚群组成较为一致,但有2个采样群体存在一定的杂交特征;主成分分析显示,3个采样群体与其他西藏山羊品种或群体在前2个主成分上分离,位置及相互关系符合进化树结构以及群体结构结果。该研究从基因组水平解释了昌都黑山羊的种质特性,也为后续藏山羊种质资源保护和利用、重要经济性状的基因定位以及分子育种提供重要的理论依据。Tibetan goat is a unique domestic breed in China.In order to further understand the genetic diversity and population genetic structure of Tibetan goats at the genomic level,whole-genome resequencing(WGS)of Changdu black goats from three different populations was carried out in this study.WGS was used to identify SNP in three local populations,and a total of 13,426,331 SNP were identified.The results of genetic diversity and linkage disequilibrium(LD)analysis showed that the nucleotide diversity of the three local populations was similar,and the LD decay curves were similar as well.Through NJ phylogenetic tree analysis,its topology showed that the three local groups clustered into one branch,which was significantly separated with other eight goat breeds in Tibet.The population structure analysis showed that the composition of subgroups among the local populations was relatively consistent,but there were certain hybridization characteristics between the two local populations.Principal component analysis showed that the three local populations were separated from other Tibetan goat breeds or populations on the first two principal components,and their positions and relationships were in line with the results of evolutionary tree structure and population structure.This study demonstrated the germplasm characteristics of the Tibetan Changdu black goat at the genomic level,and provided an important theoretical basis for the subsequent protection and utilization of Tibetan goat resources,gene mapping of important economic traits and molecular breeding.
