摘 要:猪精子冷冻是长久保存珍贵家猪品种遗传物质的有效手段,然而因其细胞膜结构组成的特殊性,猪精子经过冷冻后活力大不如前。熊果酸是一种天然的、具有抗氧化作用羧酸类化合物,在其他多个领域已被报道具有优异的抗氧化功能。为研究熊果酸对长白公猪冷冻精液的作用,该试验在猪精子冷冻稀释液中分别添加不同浓度梯度(0、1.0、1.4、1.6、1.8和2.0 mg/mL)的熊果酸,并检测冷冻-解冻后猪精子的活力及其他相关参数。结果表明,当添加1.6 mg/mL熊果酸提取液时,与对照组相比较,精子活力(50.27%±3.24%)显著增加(P<0.05);同时STR和VAP数值明显提高(分别为54.95%±3.35%和36.53±3.28μm/s)(P<0.05),VCL也最高(59.63±3.11μm/s)(P<0.05)。每个处理组之间VSL、LIN、ALH值无显著性差异(P<0.05)。结果表明,1.6 mg/mL熊果酸的添加可以显著提高猪冷冻精子的各项质量参数。Cryopreservation of boar sperm is an effective method to preserve the genetic material of domestic boar breeds for a long time.However,due to the special structure of the cell membrane,the viability of frozen boar spermatozoa is significantly lower than before.Ursolic acid,a natural antioxidant carboxylic acid compound,has been reported to have excellent antioxidant functions in many other fields.In this study,ursolic acid of different concentration gradient(0,1.0,1.4,1.6,1.8 and 2.0 mg/mL)was added to the frozen diluent of boar sperm,and the viability and other related parameters of boar sperm after freeze-thaw were detected.The results showed that when 1.6 mg/mL ursolic acid was added,the sperm motility was the most significant(50.27±3.24)compared with the control group(P<0.05).Meanwhile,STR and VAP values were significantly increased(54.95±3.35 and 36.53±3.28,respectively)(P<0.05),and VCL parameters were the highest(59.63±3.11,P<0.05).There was no significant difference in VSL,LIN,ALH and BCF values between each treatment group(P<0.05).The results showed that the addition of 1.6mg/mL ursolic acid could significantly improve the quality parameters of frozen pig sperm.
