告达庭通过调控menin/β-catenin表达抑制卵巢癌生长

摘　　要：目的研究告达庭对卵巢癌生长的影响及其机制。方法细胞实验分为4组:对照组、低剂量组(15μmol/L)、中剂量组(30μmol/L)、高剂量组(60μmol/L)告达庭作用卵巢癌细胞CAOV324 h。CCK-8检测细胞增殖;流式细胞术检测凋亡;Western blot检测相关蛋白表达。采用RNA干扰技术,抑制多发性内分泌腺瘤1型基因编码蛋白(menin)表达,观察告达庭对凋亡相关蛋白的表达作用。建立卵巢癌皮下移植瘤模型,观察瘤的生长,实时荧光定量PCR(realtime PCR)检测凋亡相关基因的表达。结果告达庭可抑制卵巢癌细胞生长,并促进其凋亡;告达庭干预后,Nuclearβ-catenin、DNA甲基转移酶1(DNMT1)、survivin及c-myc表达均减少,而menin表达增加。抑制menin表达,逆转告达庭对相关蛋白的表达作用。在体内实验中,告达庭可抑制移植瘤的生长;抑制menin表达,逆转告达庭对卵巢癌组织内相关基因表达。结论告达庭可抑制卵巢癌增殖,并促进其凋亡,机制可能与调控menin/β-catenin信号通路有关。Objective To investigate the effect and the mechanism of Caudatin on growth of ovarian cancer.Methods Theovarian cancer cells were divided into 4 groups:control group,low dose group(15μmol/L),medium dose group(30μmol/L)and high dose group(60μmol/L).Ovarian cancer cell CAOV3 was treated with Caudatin for 24 h.CCK-8 was used to detectcell proliferation.Flow cytometry was used to detect apoptosis.The expression of related proteins was detected by Western blot.RNA interference was used to inhibit the expression of multiple endocrine adenoma type 1 gene coding protein(MENIN),andto observe the effect of Caudatin on the expression of apoptosis-related proteins.The subcutaneous transplantation tumor modelof ovarian cancer was established,and the tumor growth was observed.The expression of apoptosis-related genes was detectedby real-time PCR.Results Caudatin restrains the proliferation of CAOV3 cells and increased the apoptosis.Caudatin canreduce the protein expression of Nuclearβ-catenin,DNMT1,survivin and c-myc,while increase the protein expression ofmenin.menin inhibition reverses the effect of Caudatin on apoptosis related protein expression in ovarian cancer cells.In theexperiment of transplanted tumor in vitro,Caudatin can inhibit the growth of transplanted tumor.menin inhibition reverses theeffect of Caudatin on apoptosis related genes in ovarian cancer tissue.Conclusion Caudatin can restrain the proliferation andinduce apoptosis of ovarian cancer.Its mechanism may be related to regulateβ-catenin signal pathway.
