38例成人皮肌炎患者外周血淋巴细胞亚群分布异常的临床研究

摘　　要：目的研究成人皮肌炎患者与对照组健康者外周血B淋巴细胞、T淋巴细胞及自然杀伤(NK)细胞有无差异及临床意义。方法收集浙江省人民医院2017年1月—2021年9月38例成人皮肌炎患者和38例健康体检者的1 ml外周血,采用荧光抗体标记的方法,使用20μl外周血和5μl荧光抗体共同孵育,并通过CantoII型流式细胞仪检测外周血中CD3^(+)、CD4^(+)、CD8^(+)、CD19^(+)B淋巴细胞以及CD3-CD16^(+)CD56^(+)NK细胞的分布频率,并进行统计学分析。结果38例成人皮肌炎患者CD4^(+)T淋巴细胞和NK细胞频率显著高于对照组(P<0.05),而B淋巴细胞频率显著低于对照组(P<0.05)。结论CD4^(+)T淋巴细胞和NK细胞频率异常增高在成人皮肌炎患者中可能扮演重要作用。Objective To study the difference and clinical significance of B lymphocytes,T lymphocytes and natural killer(NK)cells in peripheral blood between adult dermatomyositis patients and healthy control people.Methods The peripheral blood of 38 cases of dermatomyositis and 38 control cases from January 2017 to September 2021 in Zhejiang Provincial People.s Hospital were collected.The 20μl peripheral blood of each sample and 5μl fluorescent labeling antibody was incubated.Then the lymphocyte subsets of peripheral blood were detected by Canto II flow cytometry.Then the distribution frequency of CD19^(+)B lymphocytes and CD3^(+)-CD16^(+)CD56^(+)NK cells were statistically analyzed.Results The frequencies of CD4^(+)T lymphocytes and NK cells in 38 adult dermatomyositis patients were significantly higher than control group(P<0.05),while the frequencies of B lymphocytes were significantly lower than control group(P<0.05).Conclusion The significantly elevated CD4^(+)T lymphocytes and NK cells may play an important role in the progression of adult dermatomyositis patients.
