摘  要:目的探讨邻苯二甲酸二甲酯(DMP)对雄性小鼠睾丸标志酶基因表达水平的影响。方法以SPF级C57雄性小鼠为实验动物,经不同剂量染毒8周后,取睾丸组织提取总RNA。实时荧光定量RT-PCR技术检测睾丸组织乳酸脱氢酶(LDH1)、γ-谷氨酰转肽酶(GGT,γ-GT)、碱性磷酸酶(AKP)、葡萄糖-6-磷酸脱氢酶(G-6-PD)的基因表达水平。结果与对照组相比较,LDH1基因表达水平在DMP各剂量组差异无统计学意义(P>0.05);GGT基因表达水平在DMP各剂量组中均低于对照组,差异均有统计学意义(P<0.01);AKP基因表达水平在DMP各剂量组中均低于对照组,其中500 mg/kg剂量组比较对照组差异有统计学意义(P<0.05);G-6-PD基因表达水平在DMP各剂量组中均高于对照组,差异均有统计学意义(P<0.05)。结论DMP暴露可引起小鼠睾丸细胞标志酶对应基因表达水平的改变,影响酶的合成,可能诱发雄性小鼠生精功能障碍。Objective To investigate the effects of dimethyl phthalate(DMP)on the gene expression of Teticular marker enzymes in male mice.Methods Total RNA was extracted from the testis of SPF C57 male mice after exposure to different concentrations of DMP for 8 weeks.The gene expression of lactate dehydrogenase(LDH1),glutamyl transpeptidase(GGT),alkaline phosphatase(AKP),glucose-6-phosphate dehydrogenase(G-6-PD)in the testes was detected by real-time quantitative RT-PCR.Results Compared with control group,the gene expression of LDH1 was downregulated in DMP exposure groups(P>0.05),and the gene expression of GGT was significantly downregulated in DMP exposure groups,with the differences statistically significant(P<0.01).The expression of AKP gene in all DMP dose groups was lower than that in the control group,and the difference was statistically significant in the 500 mg/kg dose group compared with the control group(P<0.05).The expression of G-6-PD gene in DMP dose groups was higher than that in control group,and the differences were statistically significant(P<0.05).Conclusion DMP exposure in mice leads to the gene expression change of Teticular marker enzymes and interferes with the synthesis of the enzymes,which could cause spermatogenesis dysfunction in male mice.
