肺内源性和外源性急性呼吸窘迫综合征小鼠生物标志物水平的比较

摘  要:目的观察肺内和肺外源性急性呼吸窘迫综合征小鼠肺损伤标志物的差异,为临床治疗提供参考。方法将30只8周龄C57BL/6JGpt雄性小鼠随机分为外源性ARDS组、外源性NS组、内源性ARDS组、内源性NS组和对照组共五组,每组各6只。外源性和内源性ARDS组小鼠模型分别采用肺内注射和腹腔注射LPS10 mg/kg诱导建立,外源性NS组与内源性NS组则分别肺内注射和腹腔注射等量无菌生理盐水,在成功造出ARDS小鼠模型后选取24小时时间点采集肺组织标本,进行HE染色,并根据切片进行肺损伤评分,每组肺组织中Ang-2、SP-D、ESM-1的相对表达量使用qRT-PCR法来检测。结果ARDS模型组肺损伤评分升高(P<0.01),肺组织ANG-2、ESM-1、SP-D水平均升高,外源性ARDS组肺组织ANG-2、ESM-1水平高于内源性ARDS组,而SP-D水平低于内源性ARDS组。结论肺内外源性ARDS肺损伤的机制不同,外源性ARDS以破坏肺毛细血管内皮细胞为主,而内源性ARDS主要引起肺泡上皮细胞损伤。Objective To observe the differences of lung injury markers in mice with intra-pulmonary and extrinsic pulmonaryacute respiratory distress syndrome,and to provide reference for clinical treatment.Methods Thirty 8-week-old C57 BL/6 JGpt male mice were randomly divided into exogenous ARDS group,exogenous NS group,endogenous ARDS group,endogenous NS group and control group,with six mice in each group.Exogenous and endogenous ARDS mouse models were induced by intratracheal injection and intraperitoneal injection of LPS 10 mg/kg respectively.Exogenous NS group and endogenous NS group were injected withsterile normal saline through the same way.After the successful establishment of ARDS mouse model,lung tissue samples were collected at 24 hours,stained with hematoxylin eosin(HE),and lung injury scores were performed according to slice.The relative expression of ANG-2,SP-D,ESM-1 was detected by qRT-PCR.Results The lung injury score of ARDS group increased(P<0.01),and the levels of ANG-2,ESM-1 and SP-D in lung tissue were increased.The levels of ANG-2 and ESM-1 in lung tissue of exogenous ARDS group were higher than those of endogenous ARDS group,while the levels of SP-D were lower than those of endogenous ARDS group.Conclusions The mechanisms of lung injury in exogenous and endogenous ARDS are different.Exogenous ARDS mainly destroys pulmonary capillary endothelial cells,while endogenous ARDS mainly causes alveolar epithelial cell damage.
