摘  要:为探讨竞争性等位基因特异性PCR(KASP)标记在玉米DUS(特异性、一致性和稳定性)测试过程中近似品种筛选方面的应用,采用表型和KASP标记对玉米测试品种进行聚类分析。表型的二阶聚类结果显示,315份玉米品种可划分为普通型、甜质型以及糯质型和甜糯混合型3类,“籽粒:类型”性状对聚类结果的影响最大;系统聚类结果显示,单交种和自交系间的分离比较明显,普通型玉米与甜质型、糯质型、甜糯混合型玉米间的分离也较明显,甜质型、糯质型、甜糯混合型玉米三者之间的分离不明显。KASP标记聚类结果显示,自交系与单交种交织在一起,普通型玉米与甜质型、糯质型、甜糯混合型玉米间明显分离,甜质型玉米与糯质型、甜糯混合型玉米间的分离也较明显,糯质型玉米和甜糯混合型玉米相互交织。综上分析可知,在繁殖类型上单交种和自交系表型聚类结果与KASP标记聚类结果不同,而在籽粒类型上2种聚类结果基本吻合。To explore the application of kompetitive allele-specific PCR(KASP)markers in similar varieties selection in maize distinctness,uniformity and stability(DUS)test,based on the data of morphological characteristics and KASP markers,315 varieties were studied,including two breeding types of inbred lines and hybrids and four grain types of normal,sweet,waxy,sweet-waxy.The results of two-step cluster analysis showed that the four grain types were divided into three groups of normal,sweet,waxy and sweet-waxy.The effect of the grain:type trait on clustering results was the greatest.Results of hierarchical clustering indicated that inbred lines and hybrids were clearly separated.The separation of normal type and sweet type,waxy type,sweet-waxy type was relatively obvious,while the separation among sweet type,waxy type,sweet-waxy type was not obvious.The clustering results based on KASP markers showed that inbred lines were distributed in hybrids.Normal type and sweet type,waxy type,sweet-waxy type were significantly separated,sweet type and waxy type,sweet-waxy type were also clearly separated,while waxy type and sweet-waxy type blended with each other.In conclusion,the results of two clustering methods used in this study were different in the reproductive type,while the results were basically consistent in the grain type.
