化浊解毒汤对糖尿病周围神经病变小鼠神经电生理水平的影响

摘  要:目的探究化浊解毒汤对糖尿病周围神经病变(DPN)小鼠神经电生理水平的影响。方法选取20只SPF级db/db雄性小鼠(30~40 g)及10只db/m小鼠进行研究,分组:20只db/db小鼠全部造模成功后,观察无异常则按体质量顺序从高到低依次编号1~20,随后按数字排序法将20只DPN小鼠分为2组(模型组、化浊解毒汤组各10只)。对照组的10只db/m小鼠亦用此法编号。化浊解毒汤组给予灌胃化浊解毒汤,对照组及模型组灌胃给予等体积的水,每日灌胃1次,持续灌胃12周。在第6周、12周禁食12 h后,给予小鼠乙醚麻醉,取小鼠眼底静脉丛血液,处理后,利用酶标仪检测对应波长下的吸光度,检测血糖血脂指标[高密度脂蛋白胆固醇(HDL-C)、三酰甘油(TG)、总胆固醇(TC)、空腹血糖(FBG)]水平、氧化应激指标[超氧化物歧化酶(SOD)、转化生长因子-β1(TGF-β1)、核因子κB(NF-κB)及谷胱甘肽(GSH)]水平,测量痛觉阈值(小鼠舔后足反应的潜伏期),而且连接Power Lab生理记录仪测量神经电生理水平[感觉神经传导速度(SNCV)、运动神经传导速度(MNCV)]。结果治疗6周、12周后化浊解毒汤组LDL-C、FBG、TG、TC水平低于模型组(P<0.05)。与对照组相比,6周、12周后模型组小鼠SOD、TGF-β1、NF-κB水平显著提高,GSH水平降低(P<0.05;化浊解毒汤组小鼠SOD、TGF-β1、NF-κB水平较模型组低,GSH水平较模型组高(P<0.05)。化浊解毒汤组小鼠SNCV、MNCV、痛觉阈值水平较模型组高(P<0.05)。结论化浊解毒汤能够控制血糖血脂,提高神经电生理水平及痛觉阈值,其作用机制可能与改善氧化应激有关。Objective To explore the effect of Huazhuo Jiedu decoction on the level of neuroelectrophysiology in diabetic peripheral neuropathy(DPN)mice.Methods Twenty SPF DB/DB male mice(30-40 g)and 10 DB/m mice were selected for this study.Grouping:After all the 20 DB/DB mice were successfully modeled,if no abnormality was observed,they were numbered 1-20 in order of weight from high to low.Then,20 DN mice were divided into 2 groups(model group and Huazhuo Jiedu decoction group,with 10 mice in each group)by numerical sorting method.Ten DB/m mice in the control group were also numbered by this method.Huazhuo Jiedu decoction group was given Huazhuo Jiedu decoction by gavage,control group and model group were given equal volume of water by gavage once a day for 12 weeks.After fasting for 12 hours at 6w and 12w,the mice were anesthetized with ether,and the blood of the fundus venous plexus of the mice was taken for treatment.Absorbance at corresponding wavelength was detected by elisa,blood glucose and lipid indexes(LDL-C,TG,TC,FBG),oxidative stress indexes(SOD,TGF-β1,NF-κB and GSH levels)were detected,and pain thresholds were measured[the latency of foot reaction after licking in mice].The electrophysiological levels of sensory nerve conduction velocity(SNCV)and motor nerve conduction velocity(MNCV)were measured by a Power Lab physiological recorder.Results The levels of LDL-CFBG,TG and TC in Huazhuo Jiedu decoction group were lower than those in model group after 6w and 12w treatment(P<0.05).Compared with the control group,the levels of SOD,TGF-β1 and NF-κB in model group were significantly increased 6w and 12w after treatment,while the levels of GSH were decreased(P<0.05).The levels of SOD,TGF-β1 and NF-κB in Huazhuo Jiedu decoction group were higher than those in model group,while the GSH level was lower than that in model group(P<0.05).The levels of SNCV,MNCV and pain threshold in Huazhuo Jiedu decoction group were higher than those in model group(P<0.05).Conclusion Huazhuo Jiedu decoction can control blood glucose
