摘 要:目的观察鸭跖草总黄酮对原发性高血压大鼠肾素-血管紧张素-醛固酮系统(RAAS)的影响。方法将原发性高血压模型大鼠随机分为模型组、卡托普利组、鸭跖草总黄酮高剂量组、鸭跖草总黄酮低剂量组,每组10只;另设10只Wistar大鼠为正常组。各组灌胃卡托普利(40 mg/kg)、鸭跖草总黄酮(100 mg/kg、200 mg/kg),每日1次,连续12周;正常组、模型组灌胃等体积生理盐水。给药后6周、12周测量大鼠血压。末次给药2 h后,3%苯巴比妥钠腹腔注射麻醉后,腹主动脉取血,分离血清,检测肾素、醛固酮(ALD)、血管紧张素Ⅱ(AngⅡ)、血管紧张素转化酶(ACE)、一氧化氮(NO)、内皮素-1(ET-1)含量;分离胸主动脉,测定血管舒张和收缩功能。结果与模型组比较,卡托普利组、鸭跖草总黄酮各剂量组血压及血清肾素、ALD、AngⅡ、ACE、ET-1含量降低,NO含量升高,胸主动脉收缩和舒张功能紊乱有所改善(P<0.05或P<0.01)。结论鸭跖草总黄酮对高血压大鼠有较好的降血压效果,其作用可能通过改善胸主动脉收缩和舒张功能,调节血清肾素、ALD、AngⅡ、ACE、NO、ET-1含量及RAAS影响血压。Objective To observe the effect of total flavonoids of commelina communis on renin-angiotensin-aldosterone system in essential hypertensive rats.Methods The essential hypertension model rats were randomly divided into model control group,captopril group,low-dose total flavonoids of commelina communis group and high-dose total flavonoids of commelina communis group,with 10 rats in each group.The other 10 Wistar rats were set as normal group.Each experiment groups were gavaged with captopril(40 mg/kg)and total flavonoids of Commelina officinalis(100 mg/kg,200 mg/kg),once a day for 12 weeks.The normal group and the model group were gavaged with an equal volume of normal saline.The blood pressure of the rats was measured after 6th and 12th weeks administration.Two hours after the last administration,after intraperitoneal injection of 3%phenobarbital sodium,blood was collected from the abdominal aorta,serum was separated,and the contents of renin,aldosterone(ALD),angiotensinⅡ(AngⅡ),angiotensin-converting enzyme(ACE),nitric oxide(NO),and endothelin-1(ET-1)were detected.The thoracic aorta was isolated,and vasodilation and systolic function were detected in vitro.Results Compared with the model group,blood pressure and the levels of renin,ALD,AngⅡ,ACE,and ET-1 in the captopril group and Commelina flavonoids groups were decreased,NO was increased,and thoracic aortic systolic and diastolic dysfunction were improved(P<0.05 or P<0.01).Conclusion The antihypertensive effect of total flavonoids of commelina communis might be related to improve the systolic and diastolic dysfunction of the thoracic aorta,regulate the RAAS effected by renin,ALD,AngⅡ,ACE,NO,and ET-1.
