摘　　要：目的:优化生乳汤的提取工艺。方法:建立生乳汤中槲皮素和甘草酸含量测定方法,采用单因素试验,考察提取时间、水料比和提取次数3个因素对槲皮素提取量的影响。结合Box-Behnken响应面法,以槲皮素和甘草酸提取率综合评分为响应指标,考察提取时间、水料比和提取次数3个因素对提取工艺的影响。结果:槲皮素在0.27~43.52μg,甘草酸在1.01~201.20μg范围内线性关系良好(r>0.999),槲皮素和甘草酸平均加样回收率分别为101.50%、101.17%,RSD分别为1.81%、1.94%。根据回归方程确定的最佳提取工艺为:提取时间为1h,液料比为20∶1,提取次数为2次。结论:采用该工艺条件,生乳汤中槲皮素和甘草酸提取率分别为7.15%和4.10%,与理论预测值接近,为生乳汤的应用与开发提供理论依据。Objective:To optimize the extraction process of Shengru Decoction.Methods:The content determination method of quercetin and glycyrrhizic acid in Shengru Decoction was established.The effects of extraction time,liquid-solid ratio and extraction times on the extraction amount of quercetin were investigated by single factor test.Combined with Box-Behnken response surface method,the effects of extraction time,liquid-solid ratio and extraction times on the extraction process were investigated with the comprehensive score of quercetin and glycyrrhizic acid extraction rate as the response index.Results:The linear ranges of quercetin and glycyrrhizic acid were 0.27-43.52μg and 1.01-201.20μg(r>0.999),respectively.The average recoveries of quercetin and glycyrrhizic acid were 101.50%and 101.17%,with RSD of 1.81%and 1.94%,respectively.According to the regression equation,the optimum extraction process was as follows:extraction time was 1 h,liquid-solid ratio was 20∶1,extraction times was 2 times.Conclusion:The extraction rates of quercetin and glycyrrhizic acid are 7.15%and 4.10%,respectively,which are close to the predicted values.The results provide a theoretical basis for the application and development of Shengru Decoction.
