摘 要:目的:观察模拟微重力下骨碎补柚皮苷对T淋巴细胞干预下的成骨细胞BMP-2及OPN蛋白表达的影响。方法:取SD乳鼠颅盖骨提取成骨细胞进行培养,按照实验分组,与原代T淋巴细胞共培养24 h,将对照组置于微重力模型中培养,将骨碎补主要成分柚皮苷作为药物干预进行实验,其余参照组均加入同体积培养液于37℃,5%CO_(2)培养箱培养24 h,显微镜下观察成骨细胞增殖变化,ALP活性检测盒检测成骨细胞的ALP表达,通过ELISA方法和Western blot方法检测BMP-2蛋白表达;通过ELISA方法检测OPN蛋白表达。结果:与对照组相比,在模拟微重力情况下,与T淋巴细胞共培养组的成骨细胞BMP-2及OPN蛋白表达显著下降(P<0.05);加入骨碎补柚皮苷药物干预后BMP-2及OPN表达有所增加,差异有统计学意义(P<0.05)。结论:模拟微重力下骨碎补柚皮苷能够改善T细胞干预的成骨细胞的BMP-2及OPN的蛋白表达,从而促进成骨细胞的增殖及分化能力。Objective:To explore the effect of Drynaria fortunei naringin on the protein expressions of BMP-2 and OPN in osteoblasts mediated by T lymphocytes under simulated microgravity.Methods:Osteoblasts were extracted from the calvaria of SD suckling mice and cultured.They were grouped according to the experiment and co-cultured with primary T lymphocytes for 24 hours.The control group was cultured in a microgravity model which Drynaria fortunei naringin was used as a drug intervention.The remaining control groups were all added with the same amount volume of culture medium and the cells were cultured in 5%CO_(2) incubator at 37℃for 24 hours.The changes in osteoblasts proliferation were explored under the microscope.ALP expression of osteoblasts was detected by ALP activity kit.The protein expression of BMP-2 was detected by ELISA method and Western blotting method,and the protein expression of OPN was detected by ELISA method.Results:Compared with those in the control group,the protein expressions of BMP-2 and OPN decreased significantly in osteoblasts co-cultured with T lymphocytes under simulated microgravity(P<0.05);which were increased after the intervention of Drynaria fortunei naringin(P<0.05).Conclusion:Drynaria fortunei naringin can improve the protein expressions of BMP-2 and OPN in osteoblasts mediated by T cells under simulated microgravity,thereby promoting the proliferation and differentiation of osteoblasts.
