摘  要:目的采用干扰素γ(interferon-γ,IFN-γ)诱导人口腔鳞癌细胞建立体外PD-L1高表达细胞模型,并探讨IFN-γ对口腔鳞癌细胞生物学行为能力的影响。方法选取人口腔鳞癌细胞系SCC15进行培养,通过RT-PCR和流式细胞实验检测IFN-γ对SCC15细胞PD-L1 mRNA和蛋白表达的影响,然后通过MTT实验、划痕实验、Transwell实验和克隆形成实验检测IFN-γ对SCC15细胞增殖、迁移、侵袭和克隆形成能力的影响。结果IFN-γ处理人口腔鳞癌细胞SCC15后,细胞中PD-L1 mRNA和蛋白的表达水平显著上调,并呈一定程度剂量依赖性。此外,IFN-γ可以促进人口腔鳞癌细胞SCC15的迁移、侵袭和克隆形成能力,而细胞的增殖活性受到轻微抑制。结论IFN-γ能成功诱导人口腔鳞癌细胞建立体外PD-L1高表达细胞模型,同时促进人口腔鳞癌细胞的迁移、侵袭和克隆形成,并且在一定程度上抑制口腔鳞癌细胞的增殖。Objective To establish an in vitro PD-L1 high-expressing cell model in human oral squamous cell carcinoma cells by IFN-γ,and to examine the effect of IFN-γon the biological behaviors of oral squamous cell carcinoma cells.Methods The human oral squamous cell carcinoma cell line SCC15 was selected for culture,and the effect of IFN-γon the expression of PD-L1 mRNA and protein in SCC15 cells was detected by Real-time PCR and flow cytometry,and the effect of IFN-γon the proliferation,migration,invasion and colony formation ability of SCC15 cells was detected by MTT assay,scratch assay,and transwell and colony formation experiments.Results The expression levels of PD-L1 mRNA and protein in the cells were significantly up-regulated after IFN-γtreatment.In addition,IFN-γcould promote the migration,invasion and colony formation of human oral squamous cell carcinoma cells SCC15,but the proliferation activity was slightly inhibited.Conclusions An in vitro PD-L1 high expression model in human oral squamous cell carcinoma cells was successfully established through IFN-γ.IFN-γtreatment could promote the migration,invasion and colony formation of human oral squamous cell carcinoma cells,and slightly inhibit the proliferation of oral squamous cell carcinoma cells.
