摘  要:目的探讨早孕绒毛及蜕膜组织中白细胞介素6(IL-6)对吲哚胺2,3-双加氧酶(IDO)表达的影响。方法收集行人工流产终止妊娠的37例正常早孕妇女的绒毛及蜕膜组织,用Western blot法检测两种组织中IDO及信号转导及转录激活因子-1(STAT1)蛋白的表达;在早孕绒毛、蜕膜组织中加入不同浓度(0、50×10^(-6)及100×10^(-6) mg/L)的IL-6培养24 h,检测两种组织中IDO、蛋白质酪氨酸磷酸酶-1(SHP-1)、蛋白质酪氨酸磷酸酶-2(SHP-2)及STAT1蛋白的表达。结果早孕绒毛及蜕膜组织中均检测到IDO和STAT1蛋白的表达;在培养的正常早孕绒毛和蜕膜组织中,IL-6能上调IDO、SHP-1、SHP-2及STAT1蛋白的表达,且IL-6浓度与IDO、SHP-1、SHP-2及STAT1蛋白的表达呈正相关(P<0.05)。结论IL-6的浓度能影响体外培养的早孕绒毛及蜕膜组织中IDO蛋白的表达。Objective To investigate the effect of interleukin-6(IL-6)on indoleamine 2,3-dioxygenase(IDO)in villi and decidua in early pregnancy.Methods The villi and decidua tissues were collected from 37 normal pregnant women who underwent induced abortion.Western blot was used to detect the expression of IDO and signal transducerand activator of transcription 1(STAT1)in villi and decidua.Collected villi and decidua tissues were cultured under different concentrations(0,50×10^(-6) and 100×10^(-6) mg/L)of IL-6 for 24 h.The protein levels of IDO,Src-homology 2-domain phosphatase-1(SHP-1),SHP-2,and STAT1 were examined.Results The protein expression of IDO and STAT1 were detectable in the villi and decidua of early pregnancy.In cultured normal villi and decidua,IL-6 upregulated the expression levels of IDO,SHP-1,SHP-2,and STAT1.IL-6 concentration was positively correlated with the protein expression levels of IDO,SHP-1,SHP-2,and STAT1(P<0.05).Conclusion The concentration of IL-6 can affect IDO protein expression in villi and decidua in early pregnancy.
