摘　　要：目的探讨白细胞介素-10(IL-10)促进子宫内膜异位症血管生成的作用机制。方法选取2016年9月至2018年9月我院收治的60例子宫内膜异位症患者为子宫内膜异位症组;选取同期在我院妇产科接受治疗的60例患者(无子宫内膜异位症,且无影响血清中IL-10水平的其他疾病)为对照组,比较两组患者的血清IL-10水平。建立体外人脐静脉内皮细胞(HUVEC)模型,分为PBS对照组、IL-10组、白细胞介素-10受体(IL-10R)抗体组,以八肽胆囊收缩素(CCK-8)法检测细胞增殖,Transwell法检测细胞迁移,血管管腔形成实验检测细胞管腔形成能力;采用免疫印迹法检测信号转导子和转录激活子3(STAT3)、血管内皮生长因子(VEGF)及磷酸化STAT3(p-STAT3)水平并进行比较分析。结果子宫内膜异位症组的血清IL-10水平明显高于对照组(P<0.05)。IL-10以剂量依赖方式促进HUVEC细胞增殖。IL-10组的细胞侵袭能力明显高于PBS对照组、IL-10R抗体组(P<0.05)。IL-10组的管腔形成能力明显高于PBS对照组、IL-10R抗体组(P<0.05)。IL-10组细胞内VEGF及p-STAT3水平显著提高。结论IL-10通过促进细胞增殖、迁移,刺激细胞血管生成,从而诱导子宫内膜异位症的发生和发展。Objective To investigate the mechanism of interleukin-10(IL-10)promoting angiogenesis in endometriosis.Methods A total of 60 patients with endometriosis admitted in our hospital from September 2016 to September 2018 were selected as endometriosis group;a total of 60 patients(without endometriosis and other diseases affecting serum IL-10level)who were treated in the gynecology and obstetrics department of our hospital during the same period were selected as control group;the level of serum IL-10 was compared between the two groups.Human umbilical vein endothelial cells(HUVEC)model was established in vitro,which was divided into PBS control group,IL-10 group and interleukin-10receptor(IL-10R)antibody group;cell proliferation was detected by cholecystokinin-8(CCK-8)method,cell migration was detected by Transwell method,and cell lumen formation ability was detected by vascular lumen formation experiment.Western blotting was used to detect the levels of signal transducer,activator of transcription 3(STAT3),vascular endothelial growth factor(VEGF)and STAT3 phosphorylation(p-STAT3),and the above data were compared and analyzed.Results The level of serum IL-10 in the endometriosis group was significantly higher than that in the control group(P<0.05).IL-10 promoted the proliferation of HUVEC cells in a dose-dependent manner.The cell invasion ability in the IL-10 group was significantly higher than that in the PBS control group and the IL-10R antibody group(P<0.05).The lumen formation ability in the IL-10 group was significantly higher than that in the PBS control group and the IL-10R antibody group(P<0.05).The levels of VEGF and p-STAT3 significantly increased in the IL-10 group.Conclusion IL-10 induces the occurrence and development of endometriosis by promoting cell proliferation and migration and stimulating cell angiogenesis.
