猴头健胃灵片对功能性消化不良大鼠胃动力、脑肠肽和c-kit的影响

摘　　要：目的:探讨猴头健胃灵片促进功能性消化不良(FD)大鼠胃排空的可能机制。方法:将30只雄性SD大鼠随机分为空白组、模型组、猴头健胃灵组,每组10只。模型组和猴头健胃灵组采用夹尾刺激法制备FD大鼠模型。猴头健胃灵组予以猴头健胃灵片水溶液灌胃,空白组、模型组予以等体积纯水灌胃,7 d后检测大鼠胃排空率;张力换能器检测大鼠离体胃动力;酶联免疫吸附试验法检测大鼠血浆中脑肠肽胃动素(MTL)和胆囊收缩素(CCK)含量;免疫组织化学检测大鼠胃c-kit的表达水平。结果:1)与空白组比较,模型组体质量明显减轻(P<0.001),胃排空率、离体肌条动力指数显著降低(P<0.05);与模型组比较,猴头健胃灵组大鼠体质量显著增加(P<0.001),胃排空率、离体肌条动力指数明显升高(P<0.01);2)模型组大鼠血浆中脑肠肽MTL及CCK含量显著低于空白组(P<0.05),而猴头健胃灵组大鼠血浆中脑肠肽MTL及CCK水平显著高于模型组(P<0.05);3)猴头健胃灵组大鼠胃组织中c-kit表达与空白组和模型组大鼠比较均有所上调。结论:猴头健胃灵片能升高FD模型大鼠血浆中脑肠肽MTL和CCK的水平、上调胃组织中c-kit的表达水平,从而促进胃动力,这可能是其治疗FD的机制之一。Objective:To explore the potential mechanism underlying Houtou Jianweiling Tablets(HTJWL)promoting gastric motility of functional dyspepsia(FD)rats.Methods:A total of 30 male Sprague-Dawley(SD)rats were randomly divided into blank group,model group and HTJWL group,with 10 in each group.The model group and the HTJWL group were given tail clamping stimulation to establish FD model.Then the HTJWL group was administrated with HTJWL solution by gavage,while the model group and the blank group were given equal volume of pure water by gavage,once a day.After 7 days of treatment,gastric emptying rate was detected and gastric motility was also tested in vitro through force transducer.The concentrations of brain-gut peptides motilin(MTL)and cholecystokinin(CCK)in rat serum were detected by enzyme linked immunosorbent assay(ELISA),and the expression of c-kit in stomach was detected by western blot.Results:1)Compared with the blank group,the model group manifested weight loss(P<0.001)and decreased gastric emptying rate and motility index of gastric smooth muscle(P<0.05).Compared with the model group,the HTJWL group gained weight(P<0.001)and had increased gastric emptying rate and motility index of gastric smooth muscle(P<0.01).2)The contents of serum MTL and CCK in the model group was lower than those in the blank group(P<0.05),while the contents of serum MTL and CCK in the HTJWL group were higher than those in the model group(P<0.05).3)The expression of c-kit in stomach in the HTJWL group was up-regulated compared with that in the blank group and the model group.Conclusion:HTJWL could raise the concentrations of MTL and CCK in the serum of FD model rats,and increase the expression level of c-kit in stomach to promote gastric motility,which might be one of the mechanisms for its treatment of FD.
