摘 要:为获得隐秘小环藻(Cyclotella cryptica)的无菌藻株,本研究开展了隐秘小环藻的药敏实验及抗生素耐受性实验,初步确定了除菌抗生素的种类及浓度,并对抗生素处理前后藻液的细菌多样性和群落结构进行了对比分析,在此基础上,设计了有效的抗生素组合进行除菌。研究表明,800μg·mL^(-1)硫酸卡那霉素、800μg·mL^(-1)硫酸链霉素、600μg·mL^(-1)硫酸庆大霉素抑菌效果较好且能够显著促进隐秘小环藻生长,采用以上三种抗生素的组合能去除藻液中大部分细菌,离心洗涤后,再追加25μg·mL^(-1)盐酸环丙沙星可去除残留的Muricauda菌,实现隐秘小环藻的无菌化培养。研究结果表明,在对微藻进行无菌化处理时,抗生素的种类、浓度和添加方式是获得无菌藻株的关键,研究结果可为微藻的无菌化处理提供参考。In order to obtain the axenic Cyclotella cryptica,we carried out the susceptibility and tolerance tests of Cyclotella cryptica,and then the types and concentrations of antibiotics for removing bacteria were preliminarily determined.Bacterial diversity and community structure in cultured fluid of Cyclotella cryptica before and after antibiotic treatment were compared and analyzed.On this basis,an effective antibiotic combination was designed for sterilization.The research showed that the concentration of 800μg·mL^(-1)kanamycin sulfate,800μg·mL^(-1)streptomycin sulfate and 600μg·mL^(-1)gentamicin sulfate was more effective in inhibiting bacteria and significantly promoted the growth of Cyclotella cryptica.The combination of the above three antibiotics could remove most of the bacteria in cultured fluid of Cyclotella cryptica,after centrifugal washing,and additional 25μg·mL^(-1)ciprofloxacin hydrochloride could remove the residual bacteria Muricauda,realizing the axenic culture of Cyclotella cryptica.In summary,the types and concentrations and the addition method of antibiotic are the keys to obtain sterile microalgae strains during the axenic treatment of microalgae,which provided an important reference for the axenic treatment of microalgae.
