摘  要:目的 研究环裂松萝干燥地衣体的化学成分及抗菌活性。方法 采用开放柱色谱和制备液相进行分离纯化,根据波谱数据鉴定化合物结构。通过纸片扩散法初步筛选抗金黄色葡萄球菌的活性成分,并通过分子对接青霉素结合蛋白PBP2a虚拟筛选抗耐甲氧西林金黄色葡萄球菌(MRSA)的活性成分。结果 分离得到15个化合物,分别鉴定为扁枝衣酸乙酯(1)、扁枝衣酸(2)、atraric acid(3)、α-亚麻酸甲酯(4)、2’-O-methylevernol(5)、α-亚麻酸(6)、亚油酸(7)、苔色醛(8)、3-羟基-5-甲基苯基-2-羟基-4-甲氧基-6-甲基苯甲酸酯(9)、苔色酸(10)、瘤网地衣素(11)、苔黑酚(12)、5-羟甲基糠醛(13)、4-(乙酰氨基)丁酰胺(14)、尿嘧啶核苷(15)。化合物5、8、9在128.0 μg·mL^(-1)质量浓度下,抑菌圈直径分别为6.4、6.6、6.6 mm。分子对接结果显示,化合物6、7、11与PBP2a对接打分值较高(>40)。结论 化合物4、5、8、13~15为首次从松萝属植物中分离得到。化合物5、8、9对金黄色葡萄球菌具有一定的抑制作用;化合物6、7、11 可能具有抗MRSA的活性。Objective To determine the chemical constituents and antibacterial activity of lichen Usnea diffracta.Methods The ethanolic extract from lichen Usnea diffracta was isolated and purified by column chromatography and preparative RP-HPLC.The chemical structures were identified based on their spectroscopic data.The active ingredients against Staphylococcus aureus were screened by paper diffusion method.The active anti-MRSA ingredients were screened by molecular docking with penicillin-binding protein 2a (PBP2a).Results Totally 15 compounds were isolated and identified as ethyl everninate (1),everninic acid (2),atraric acid (3),α-linolenic acid methyl ester (4),2’-O-methylevernol (5),α-linolenic acid (6),linoleic acid (7),orsellinaldehyde (8),3-hydroxy-5-methylphenyl-2-hydroxy-4-methoxy-6-methylbenzoate (9),orsellinic acid (10),lecanorin (11),orcinol (12),5-hydromethylfurfural (13),4-(aylamino) butyramides (14),and uridine (15).Compounds 5,8,9 showed the inhibition circle of 6.4 mm,6.6 mm and 6.6 mm respectively,against Staphylococcus aureus at 128.0 μg·mL^(-1).Molecular docking showed that compounds 6,7,11 had high scoring values (>40) for docking with PBP2a.Conclusion Compounds 4,5,8,and 13-15 have been isolated from genus Usnea for the first time.Compounds 5,8,and 9 have inhibitory effect on Staphylococcus aureus.Compounds 6,7,and 11 might have anti-MRSA activity.
