摘 要:目的 采用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱(UHPLC-Q-Exactive-Orbitrap-MS)快速表征和识别苦黄注射液中的化学成分。方法 采用纯乙醇沉淀苦黄注射液后的上清液,利用UHPLC-Q-Exactive-Orbitrap-MS对样品进行定性分析,采用Waters ACQUITY UPLC HSS T3色谱柱(2.1 mm×100 mm,1.8 μm),流动相0.1%甲酸水溶液-乙腈,梯度洗脱,流速0.35 mL·min^(-1),柱温35℃,进样量5 μL,采用正、负离子监测模式。根据Xcalibur软件的精确相对分子质量、准分子离子峰、碎片离子,相关文献和数据库对苦黄注射液中的化学成分进行识别和鉴定。结果 从苦黄注射液的醇提液中共鉴定出123个化学成分,包括黄酮类、酚酸类、生物碱类、蒽醌类、萜类、氨基酸和核苷类等各种化合物。结论 首次采用UHPLC-Q-Exactive-Orbitrap-MS对苦黄注射液的化学成分进行了研究,为苦黄注射液的药效物质基础及作用机制研究提供了理论依据。Objective To rapidly characterize and identify the components of Kuhuang injection with UHPLC-Q-Exactive-Orbitrap-MS.Methods The supernatant of Kuhuang injection was precipitated by ethanol.The samples were analyzed by UHPLC-Q-Exactive-Orbitrap-MS comprehensively.Waters ACQUITY UPLC HSS T3 column (2.1 mm×100 mm,1.8 μm) was used for gradient elution with 0.1% formic acid-acetonitrile as the mobile phase in both positive and negative ion modes.The flow rate was 0.35 mL·min^(-1),the column temperature was 35℃ and the injection volume was 5 μL.The components were identified according to the accurate relative molecular weight,excimer ion peak,fragmentation in Xcalibur software and other information in literature data and database.Results Totally 123 components were rapidly identified,including flavonoids,phenolic acids,alkaloids,anthraquinones,terpenoid saponins,amino acids and nucleosides,etc.Conclusion The chemical components in Kuhuang injection are determined by UHPLC-Q-Exactive-Orbitrap-MS for the first time,which provides the theoretical basis for the study of pharmacodynamic materials basis and mechanism of Kuhuang injection.
