摘  要:目的 制备紫草多糖脂质体,优化其处方工艺,并对其进行表征。方法 逆向蒸发法制备紫草多糖脂质体,采用超速离心-紫外分光光度法测定其包封率,以包封率为主要考察指标,研究膜材比、药脂比、超声时间等因素对脂质体包封率的影响;通过正交试验对处方进行优化,并进行质量评价。结果 紫草多糖脂质体优化后的制备处方工艺为药脂比1∶4,膜材比1∶8,超声持续时间2 min,包封率为(80.62±1.09)%,平均粒径为(131.91±2.38)nm,PDI为(0.134±0.012),Zeta电位为(-18.13±0.14)mV,4℃放置10 d无沉淀生成。结论 优选处方稳定可行,制备的紫草多糖脂质体包封率高、粒径小且均匀。Objective To prepare Arnebiae Radix polysaccharide liposomes,and to characterize and optimize the formulation of Arnebiae Radix polysaccharide liposomes.Methods Arnebiae Radix polysaccharide liposomes were prepared by reverse evaporation method and the entrapped efficiency was measured by ultracentrifugation method.Single factors such as the ratio of cholesterol to phospholipid,the ratio of drug to phospholipid,and the ultrasonic time were studied.And the orthogonal design was adopted to obtain the optimized prescription,and the quality was evaluated.Results The best prescription was as follows:the ratio of drug to phospholipid of 1∶4,the ratio of cholesterol to phospholipid was 1∶8,and the ultrasonic time was 2 min.The entrapped efficiency of Arnebiae Radix polysaccharide liposomes prepared according to the prescription was (80.62±1.09)%,the average particle size was (131.91±2.38) nm,the PDI was (0.134±0.012),and the Zeta potential was (-18.13±0.14) mV.Conclusion The Arnebiae Radix polysaccharide liposomes prepared according to the optimized formulation have higher entrapped efficiency with small and uniform particle size.
