摘 要:目的 研究蒲公英提取物对白念珠菌(Candida albicans)的抑制作用及机制。方法 纸片抑菌法研究蒲公英提取物对白念珠菌的抑制作用;分光光度计测定蒲公英提取物最小抑菌浓度(MIC)并研究其抑菌动力学和细胞膜通透性;利用扫描电镜、核酸吸光度法研究蒲公英提取物对白念珠菌的抑菌机制。结果 蒲公英提取物对白念珠菌有良好的抑制作用,其MIC为32.0 mg·mL^(-1)。抑菌动力学曲线分析表明蒲公英提取物主要作用于白念珠菌的对数生长期。扫描电镜发现蒲公英提取物可破坏白念珠菌的细胞结构,导致细胞表面凹陷,形成褶皱。核酸吸光度法结果发现蒲公英提取物可以增加白念珠菌细胞膜的通透性,从而抑制其生长。结论 蒲公英提取物对白念珠菌有较好的抑制效果,该抑制作用可能是其破坏细胞壁进而导致菌体内容物外泄,从而破坏菌体细胞膜结构,对细胞增长产生抑制作用,最终导致细胞死亡。Objective To determine the inhibition effect of dandelion extract on Candida albicans(C.albicans) and related mechanism.Methods The inhibition effect of dandelion extract on C.albicans was studied by paper bacteriostasis method.The minimum inhibitory concentration(MIC) and antibacterial kinetics of dandelion extract were analyzed by spectrophotometer.On this basis,the cell membrane permeability of C.albicans was also studied.The bacteriostatic mechanism of dandelion extract on C.albicans was determined by scanning electron microscopy analysis and the UV absorption of nucleic acid method.Results The dandelion extract had a good bacteriostasis on C.albicans,with MIC of 32.0 mg·mL^(-1).The bacteriostatic kinetic curve analysis showed that the inhibition on C.albicans mainly worked in its logarithmic growth phase.Scanning electron microscopy showed that dandelion extract destroyed the cell membrane structure of C.albicans,and caused the cell surface to sink and fold.The UV absorption of nucleic acid showed the dandelion extract increased the membrane permeability of C.albicans and therefore inhibited its growth.Conclusion The dandelion extract has obvious bacteriostasis on C.albicans,which destroys the cell membrane structure,promotes the leakage of cell content,and eventually inhibits cell growth resulting in cell death.
