糖尿病肾病大鼠PLA2R与ox-LDL抗原抗体研究

摘　　要：目的:研究糖尿病肾病大鼠肾组织与血清中磷脂酶A2受体(PLA2R)与氧化低密度脂蛋白(ox-LDL)的表达与诊断价值。方法:将50只SD雄性大鼠随机分为对照组(N组)10只、模型组(M组)40只,M组适应性喂养7天,造模前禁食12 h,大剂量腹腔注射STZ(60 mg/kg),3天后测大鼠尾尖血糖≥16.7 mmol/L,即为糖尿病模型,经测定尿微量白蛋白(mALB)>30 mg/L后确定为糖尿病肾病大鼠模型成立,10周末酶联免疫法测大鼠PLA2R、ox-LDL、mALB及血糖。结果:M组大鼠PLA2R、ox-LDL表达明显高于N组,差异有统计学意义(P﹤0.01);M组大鼠肾组织及血清中PLA2R、ox-LDL与mALB呈正相关,差异有统计学意义(P﹤0.01);M组大鼠肾组织中PLA2R表达与血糖值呈正相关(P﹤0.01),血清中PLA2R表达与血糖值呈正相关(P﹤0.05);而M组大鼠肾组织及血清中ox-LDL与血糖值相关性分析无统计学意义(P﹥0.05);M组血糖值与mALB表达相关性分析无统计学意义(P﹥0.05)。结论:PLA2R、ox-LDL对糖尿病肾病有一定的临床诊断价值,且能反映病情程度。Objective:To research the expression and diagnostic value of phospholipase A2 receptor(PLA2R)and oxidized low density lipoprotein(ox-LDL)in kidney tissue and serum of rats with diabetes nephropathy.Methods:50 male SD rats were randomly divided into control group(N Group)with 10 rats and model group(M group)with 40 rats.M group was fed adaptively for 7 days,fasted for 12 hours before modeling,and injected with STZ(60 mg/kg)in large dose intraperitoneally.After three days,the blood glucose at the tip of rat’s tail was measured to be≥16.7 mmol/L,which was the diabetes model.After determining urinary microalbumin(mALB)>30 mg/L,the establishment of rat model of diabetes nephropathy was confirmed.At the end of the 10th week,PLA2R,ox-LDL,mALB and blood sugar of rats were detected by enzyme-linked immunosorbent assay.Results:The expressions of PLA2R and ox-LDL of rats in M group was significantly higher than those in N group,and the difference was statistically significant(P<0.01).PLA2R and ox-LDL in renal tissue and serum of rats in M group were positively correlated with mALB,and the difference was statistically significant(P<0.01).The expression of PLA2R in renal tissue of rats in M group was positively correlated with blood glucose(P<0.01),and the expression of PLA2R in serum was positively correlated with blood glucose(P<0.05);while the correlation analysis between ox-LDL and blood glucose value in kidney tissue and serum of rats in M group was not statistically significant(P>0.05);the correlation anal ysis between blood glucose value and mALB expression in M group was not statistically significant(P>0.05).Conclusion:PLA2R and ox-LDL have certain clinical diagnostic value for diabetes nephropathy,and can reflect the degree of the disease.
