摘 要:目的:基于“肝肠同治”理论探讨软肝散加味方对猪血清诱导肝纤维化大鼠肠道菌群的影响。方法:猪血清腹腔注射法建立肝纤维化模型。实验分为正常对照组,模型组,软肝散加味方高、中、低剂量组,每组12只,正常组、模型组予生理盐水灌胃,软肝散加味方高、中、低剂量组予软肝散加味方(20.7、41.4、82.8 g/kg)灌胃,每日1次,连续8周。HE染色观察各组大鼠肝组织病理学改变;鲎试剂法检测各组大鼠肠道内毒素含量,利用Illumina Miseq平台对大鼠16S rRNA V3-V4区的肠道菌群进行测序。结果:各给药组肝脏病理变化均有不同程度的改善,其中以软肝散加味方高剂量组改善最为明显;软肝散加味方高剂量组与中剂量组大鼠肠道内毒素含量显著降低;对造模大鼠OTUs数量、α与β多样性、菌群生物丰富度、多样性以及差异菌门、菌属的分布结构的改变均产生调整作用。结论:软肝散加味方可能通过减少内毒素生成、调节肝纤维化大鼠肠道菌群结构发挥抗肝纤维化作用。Objective:Based on the theory of“simultaneous treatment of liver and intestines”,this study investigated the effect of modified Ruangan Powder on intestinal flora in liver fibrosis rats.Methods:The liver fibrosis model was established by intraperitoneal injection of pig serum in clean male SD rats.The experiment was divided into normal control group,model group,high,medium and low dose groups of Ruangan Powder modified formula,with 12 rats in each group.The normal group and model group were given normal saline by gavage,and the high,medium and low dose groups of Ruangan Powder modified formula were given Ruangan Powder modified formula(20.7,41.4,82.8 g/kg)by gavage once a day for 8 weeks.HE staining was used to observe the pathological changes of liver tissue in each group;The intestinal endotoxin content of rats in each group was detected by limulus amebocyte lysate method,and the intestinal flora in the v3-v4 region of 16S rRNA of rats was sequenced by Illumina miseq platform.Results:The pathological changes of liver were improved in different degrees in each Ruangan Powder modified formula group,and the improvement was most obvious in the high-dose group of Ruangan Powder modified formula;The intestinal endotoxin content of rats in the high dose group and the middle dose group of Ruangan Powder modified formula was significantly decreased;The number of OTUs α And β Diversity,biological richness and diversity of flora,as well as the distribution structure of different phyla and genera in Ruangan Powder modified formula groups showed a callback effect.Conclusion:Modified Ruangan Powder may play an anti fibrosis role by reducing the production of endotoxin,regulating the structure of intestinal flora in rats with liver fibrosis.
