香诃止泻汤对功能性消化不良小鼠治疗作用的实验研究

摘　　要：目的:观察香诃止泻汤对功能性消化不良(FD)小鼠的治疗作用,并探讨其部分作用机制。方法:利用碘乙酰胺蔗糖混合溶液+游泳+左旋精氨酸溶液方法制备FD模型小鼠。将造模成功的FD腹泻小鼠随机分为模型组,香诃止泻汤高、中、低剂量组和阳性对照组,每组12只,另设正常对照组12只。香诃止泻汤高、中、低剂量组分别按33.30、16.65、8.33 g/kg给予香诃止泻汤灌胃;阳性对照组予枸橼酸莫沙必利1.95 mg/kg灌胃;模型对照组和正常对照组灌胃等量生理盐水;各组小鼠每天灌胃1次,连续灌胃3 d。观察小鼠活动情况、饮食量、体质量增长、胃内残留率、小肠推进率,ELISA法检测胃动素(MTL)、血管活性肠肽(VIP)等指标。结果:与模型组比较,各给药组平均进食量、体质量明显增加,粪便含水量、胃内残留率明显降低,小肠推进率升高,MTL水平显著升高,VIP显著降低,差异均有统计学意义(P<0.05)。结论:香诃止泻汤对FD模型小鼠具有良好的综合治疗作用,不仅能减轻FD小鼠的腹泻症状,还能增加其进食量和体质量,改善其一般状态,机制可能与调节血清MTL和VIP水平有关。Objective:To observe the therapeutic effect of Xianghe Zhixie Decoction on mice with functional dyspepsia,and explore its partial mechanism.Methods:FD models were made by using iodoacetamide sucrose mixed solution+swimming+L-arginine solution method.The successfully modeled FD diarrhea mice were randomly divided into model group,high,medium and low dose groups of Xianghe Zhixie Decoction and positive control group,with 12 mice in each group,and 12 mice in normal control group.The high,medium and low dose groups of Xianghe Zhixie Decoction were given Xianghe Zhixie Decoction by gavage at 33.30 g/kg,16.65 g/kg and 8.33 g/kg respectively;the positive control group was given Mosapride citrate 1.95 mg/kg by intragastric administration;model control group and normal control group were intragastrically administered the same volume normal saline;mice in each group were intragastrically administered once a day for 3 consecutive days.observing the changes of the mouse activity,diet,weight change,gastric residual rate,small intestinal propulsion rate,motilin,vasoactive intestinal peptide were determined by ELISA.Results:Compared with the model group,the average food intake and body weight of each medication group were significantly increased,the fecal water content and the residual rate in the stomach were significantly reduced,the small intestinal propulsion rate was increased,the MTL level was significantly increased,and the VIP level was significantly decreased,and the differences were statistically significant.Conclusion:Xianghe Zhixie Decoction has a good comprehensive therapeutic effect on FD model mice.It can not only reduce the diarrhea symptoms of FD mice,but also increase their food intake,promote their weight gain,and improve their general state.The mechanism may be related to regulating serum levels of MTL and VIP.
