摘  要:目的:研究芪风固表颗粒对肺炎链球菌(Spn)感染小鼠肺组织Toll样受体4(TLR4)/核因子-κBp65(NF-κBp65)信号通路影响。方法:实验分为正常对照组、模型组、阳性对照组(泛福舒0.63 mg/kg)和芪风固表颗粒高(2.6 g/kg)、中(1.3 g/kg)、低(0.65 g/kg)剂量组,每组10只。各药物组预防给药40 d后,乙醚麻醉小鼠,滴鼻感染7.66×10^(14)CFU/mLSpn50μL 1次,感染48 h后处死小鼠。HE染色观察各组小鼠肺组织病理变化,免疫组化检测肺组织TLR4和NF-κBP65蛋白表达水平,RT-PCR检测肺组织TLR-4和NF-κBp65mRNA相对表达水平。结果:HE染色结果表明,模型组小鼠肺组织有明显炎性浸润,芪风固表颗粒高、中剂量组肺组织炎性浸润程度减轻且支气管病变发生率低于模型组。与模型组比较,泛舒福组、芪风固表颗粒高、中剂量组肺组织TLR4和NF-κBP65蛋白表达降低(P<0.05,P<0.01),各剂量组TLR-4mRNA相对表达水平降低(P<0.01),芪风固表颗粒高剂量组NF-κBp65mRNA相对表达水平降低(P<0.05)。结论:芪风固表颗粒预防给药可减轻肺炎链球菌引起的小鼠肺部炎症,机制可能与抑制TLR4/NF-κB信号有关。Objective:To study the effects of Qifeng Gubiao Granules on Toll-like receptor 4(TLR4)/nuclear factor-κBp65(NF-κBp65)signaling pathway in the lung tissue of mice infected with Streptococcus pneumoniae(Spn).Methods:Experiment was divided into normal,model,positive(Broncho-Vaxom,0.63 mg/kg)and Qifeng Gubiao Granules high(2.6 g/kg),middle(1.3 g/kg)and low(0.65 g/kg)dose groups with each 10 mice,each group was given the corresponding medicine or saline for 40 days,then the mice were anesthetized with ether and intranasally infected with 7.66×10^(14) CFU/mL Spn 50μL once,and the mice were sacrificed 48 h after infection.HE staining was used to observe the pathological changes in lung tissue of mice in each group;immunohistochemistry was used to detect the protein expression levels of TLR4 and NF-κBp65 in lung tissue,and RT-PCR was used to detect the relative expression levels of TLR-4 and NF-κBp65 mRNA in lung tissue.Results:HE staining results showed that there was obvious inflammatory infiltration in the lung tissue of the mice in the model group.The degree of inflammatory infiltration in the lung tissue of the Qifeng Gubiao Granules high and medium dose groups was reduced and the incidence of bronchial lesions was lower than that of the model group.Compared with the model group,the expressions of TLR4 and NF-κBP65 protein in the lung tissue of the positive group,the high and medium dose groups of Qifeng Gubiao Granules were decreased(P<0.05,P<0.01),and the relative expression level of TLR-4 mRNA in each dose group was decreased(P<0.01),the relative expression level of NF-κBp65mRNA in the high-dose Qifeng Gubiao Granules group was decreased(P<0.05).Conclusion:Qifeng Gubiao Granules can improve the pulmonary inflammation caused by Streptococcus pneumoniae in mice,and the mechanism may be related to the inhibition of TLR4/NF-κB signaling pathway.
