摘  要:建立一种改良法精准控制线栓插入位置和深度,提高大鼠脑缺血再灌注(MCAO)模型成功率,为脑卒中治疗药物评价提供有效工具。SD雄性大鼠24只,分为MCAO A组(n=6),MCAO B组(n=12)和假手术组(n=6),A组将线栓全部插入,B组控制线栓插入至第二标记点。以行为学评分、水迷宫实验、TTC染色测定脑梗死面积及TUNEL染色作为评价指标,假手术组作为对照组,通过比较分析,确定建模参数以及操作要点。结果表明:该方法建模成功率为91.67%,TTC染色梗死面积为29.97%±4.46%,TUNEL染色凋亡细胞占比28.68%,是一种可靠、简便的造模方法。To improve the success rate of the middle cerebral artery occlusion(MCAO)model of SD rats,a modified method is proposed by controlling the site and the depth of suture insertion,which would provide an effective model for the treatment of cerebral stroke.24 SD male rats are divided into MCAO group A(n=6),MCAO group B(n=12)and sham-operated group(n=6).In which,the suture inserted completely is served as group A,while the suture inserted accurately on the second marker point is for group B.The survival rate,score of mNSS,Morris water maze,TTC and TUNEL staining are carried out to evaluate the MCAO model.The sham-operated group is served as the control group.After comparative analysis,the modeling parameters as well as operational points are summarized.It indicates that the modified method is a reliable simply operation as results of that the modelling success rate is increased to 91.67%,the infarct area stained by TTC is 29.97%±4.46%and the percentage of apoptotic cells stained by TUNEL is 28.68%by accurately controlling the site and the depth of the suture insertion in the method.
