摘 要:探究猪苓多糖低、中、高剂量(100 mg/kg、200 mg/kg、400 mg/kg)对良性前列腺增生(benign prostatic hyperplasia,BPH)大鼠的一般状态、前列腺指数、超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(Glutathione peroxidase,GSH-Px)、丙二醛(Malondialdehyde,MDA)水平的影响。方法为:SPF级SD大鼠70只,随机分组,其中空白组15只、造模组55只,采用腹腔注射丙酸睾丸酮建立大鼠BPH模型,造模4周后,空白组和造模组随机各选取5只大鼠处死。明确造模成功后,将造模组50只大鼠随机分为模型对照组、特拉唑嗪组和猪苓多糖低、中、高剂量组,每组各10只,经药物干预6周后处死。摘取前列腺组织计算前列腺指数(PI),考马斯亮蓝法测定蛋白质(SOD、GSH-PX)含量,MDA含量参照测试盒说明书采用比色法进行测定。结果显示:特拉唑嗪组和猪苓多糖低、中、高剂量组的前列腺指数较模型组明显降低(P<0.01),其中猪苓多糖高剂量组对前列腺增生的改善效果最为显著,其在减轻BPH前列腺组织湿重、降低BPH前列腺指数方面疗效良好。模型组BPH大鼠血清SOD、GSH-PX水平较空白组显著降低(P<0.01),MDA显著升高(P<0.01);特拉唑嗪组和猪苓多糖低、中、高剂量组SOD、GSH-PX水平高于模型组(P<0.05或P<0.01),MDA水平则显著低于模型组(P<0.01)。结论:猪苓多糖高剂量可减轻BPH前列腺组织湿重、降低BPH前列腺指数;特拉唑嗪和猪苓多糖均可提高SOD/GSH-Px,降低MDA水平,延缓BPH,以高剂量猪苓多糖效果显著。To explore the effects of low,medium and high doses(100mg/kg,200mg/kg,400mg/kg)of Polyporus umbellatus polysaccharide on the general state,prostate index,superoxide dismutase(SOD)of rats with benign prostatic hyperplasia(BPH).(SOD),Glutathione peroxidase(GSH-Px)and Malondialdehyde(MDA).Methods:Seventy SPF SD rats were randomly divided into blank group(15 rats)and model group(55 rats).The BPH model of rats was established by intraperitoneal injection of testosterone propionate.After 4 weeks,5 rats in blank group and model group were randomly selected and executed.After the success of modeling,50 rats in the model group were randomly divided into model control group,terazosin group,low,middle and high dose groups of Polyporus umbellatus polysaccharide,with 10 rats in each group,and were killed after 6 weeks of drug intervention.The prostate tissue was harvested to calculate the prostate index(PI),the content of protein(SOD,GSH-PX)was determined by Coomassie brilliant blue method,and the content of MDA was determined by colorimetry according to the instructions of test box.Results:Compared with the model group,the prostate index of terazosin group and low,middle and high doses of Polyporus umbellatus polysaccharide group decreased significantly(P<0.01),among which the high dose of Polyporus umbellatus polysaccharide group had the most significant improvement effect on prostatic hyperplasia,and it had a good effect on reducing the wet weight of BPH prostate tissue and BPH prostate index.Compared with the blank group,the serum SOD and GSH-PX levels of BPH rats in the model group were significantly lower(P<0.01),and the MDA levels were significantly higher(P<0.01).The levels of SOD and GSH-PX in terazosin group and low,middle and high doses of Polyporus umbellatus polysaccharide group were higher than those in model group(P<0.05 or P<0.01),but the level of MDA was significantly lower than that in model group(P<0.01).Conclusion:High dose of Polyporus umbellatus polysaccharide can reduce the wet weight of BPH prostate ti
