摘　　要：为了分析牦牛SOCS3基因的理化性质,本研究利用RT-PCR扩增得到牦牛SOCS3基因编码区,将其翻译成对应的蛋白质氨基酸序列后,利用生物信息学初步分析了其分子特征。结果表明,牦牛SOCS3基因编码区包含690个核苷酸,编码229个氨基酸,与普通牛、水牛、大羚羊、绵羊、骆驼、野猪、虎、大熊猫和人的SOCS3基因核苷酸序列一致性分别为99.71%、99.57%、98.70%、98.26%、98.55%、94.20%、92.75%、92.03%和91.59%。生物信息学预测表明,牦牛SOCS3蛋白属于亲水蛋白,分子式C_(1118)H_(1750)N_(310)O_(338)S_(6),预计平均分子量25.13 KD,理论等电点8.97,消光系数0.693,脂肪族系数73.23,主要分布在细胞核和细胞质,也分布在线粒体、细胞骨架等,具有潜在的磷酸化位点33个。在结构上,牦牛SOCS3蛋白含有SH2、SOCS盒结构域,二级结构主要以无规则卷曲、α-螺旋为主,还含延伸链和β-转角。本研究结果为进一步研究牦牛SOCS3提供数据信息。To analyze the physical and chemical property of yak SOCS3,we cloned the coding region of yak SOCS3 using RT-PCR and analyzed the features of SOCS3 based on bioinformatics.Our results showed that the coding region of yak SOCS3 contained 690 nucleotides,which encoded 229 amino acids,and the nucleotide homology of yak SOCS3 compared to Bos taurus,Bubalus bubalis,Oryx dammah,Ovis aries,Capra hircus,Sus scrofa,Panthera tigris,Ailuropoda melanoleuca and Homo sapiens were 99.71%、99.57%,98.70%,98.26%,98.55%,94.20%,92.75%,92.03%and 91.59%,respectively.By bioinformatic analysis,the yak SOCS3 protein was a hydropathic protein,and the molecular formula was C_(1118)H_(1750)N_(310)O_(338)S_(6),with a estimated average molecular weight of 25.13 KD,a theoretical pI of 8.97,an extinction coefficients of 0.693 and an aliphatic index of 73.23.It is mainly distributed in the nucleus and cytoplasm,and also located on mitochondria,cytoskeleton,with 33 potential phosphorylation sites.The SOCS3 protein included SH domains,and SOCS box.The second structure of yak SOCS3 was mainly random coil andα-helix,and also contained extended strand andβ-turn.Our results provide basic data information for further research on yak SOCS3.
