摘 要:为明确引起尤溪县杉木炭疽病的病原菌种类,采用常规组织分离法对采自西城镇东村村杉木人工林的疑似炭疽病病样进行病原菌分离与培养,利用形态学观察和系统发育分析对获得的病原菌株进行鉴定,同时对病原菌进行活体条件下的致病性测定。结果表明:共分离获得5株病原菌,菌落形态相似,气生菌丝白色、絮状,分生孢子无色透明,呈圆柱状,两端钝圆。基于ITS和GAPDH基因构建系统发育树,菌株S1、S3、S5、S7、S9与沧源炭疽菌Colletotrichum cangyuanense均聚在同一分支。结合形态学特征、系统发育分析和柯赫氏法则验证结果,将尤溪县杉木炭疽病病原菌鉴定为沧源炭疽菌C.cangyuanense。To recognize the pathogen causing anthracnose of Chinese fir in Youxi County, the diseased samples with typical symptoms of anthracnose were collected from the Chinese fir plantation in Dongcun village for pathogen isolation, identification and pathogenicity determination. Five isolates were systematically identified through morphological characteristics, pathogenicity test and phylogenetic analysis. The five purified isolates showed similar colonial morphology and white cotton-like hyphae. The conidia were colorless, hyaline, cylindrical with obtuse ends. The phylogenetic tree based on ITS and GAPDH genes indicated that selected isolates(S1、S3、S5、S7、S9) and Colletotrichum cangyuanense were clustered in the same branch. Based on the results of morphological characteristics, phylogenetic analysis and Koch’ s postulates, the pathogen causing anthracnose of Chinese fir in Youxi County was identified as C. cangyuanense.
