益母草碱调控LINC00461抑制宫颈癌细胞增殖并诱导细胞凋亡的实验研究

摘　　要：目的:研究益母草碱对宫颈癌细胞增殖和凋亡的影响及潜在的分子机制。方法:采用细胞计数试剂盒法和克隆形成实验检测SiHa细胞增殖,流式细胞术检测SiHa细胞凋亡率,实时荧光定量PCR检测LINC00461相对表达量,蛋白免疫印迹法检测p21和半胱氨酸蛋白酶3(Caspase-3)蛋白表达水平。结果:与未经过益母草碱处理的细胞相比,益母草碱浓度为10、20、30μg·mL^(-1)均可降低宫颈癌SiHa细胞的存活率和克隆形成数,提高细胞凋亡率,促进p21和Caspase-3蛋白表达量(P<0.05).益母草碱(30μg·mL^(-1))可显著降低细胞中LINC00461的相对表达量(P<0.05);过表达LINC00461可促进SiHa细胞增殖并抑制细胞凋亡(P<0.05);过表达LINC00461可逆转益母草碱对SiHa细胞增殖和凋亡的作用(P<0.05)。结论:益母草碱可抑制宫颈癌细胞SiHa增殖,促进细胞凋亡,其机制与抑制LINC00461表达有关。Objective:To investigate the effect of leonurine on proliferation and apoptosis of cervical cancer cells and the underlying mechanism.Methods:The proliferation of SiHa cells was detected by cell counting kit assay and clone formation experiment.The apoptosis rate of SiHa cells was detected by flow cytometry.The relative expression level of LINC00461 in SiHa cells was detected by real-time fluorescence quantitative PCR,and the expression levels of p21 and Caspase-3 proteins were detected by Western blot.Results:Compared with the cells without leonurine treatment,leonurine(10,20,30 μg·mL^(-1))could reduced the survival rate and clone number,increased the apoptosis rate,promoted the expression of p21 and Caspase-3 proteins of SiHa cells(P<0.05).Leonurine(30 μg·mL^(-1))could significantly reduced the relative expression level of LINC00461 in SiHa cells(P<0.05).Over-expression of LINC00461 could promote the proliferation and inhibit the apoptosis of SiHa cells(P<0.05).Over-expression of LINC00461 could reverse the effect of leonurine on proliferation and apoptosis of SiHa cells(P<0.05).Conclusion:Leonurine could inhibit the proliferation and promote the apoptosis of cervical cancer cells SiHa by inhibiting the expression ofLINC00461.
