摘  要:紧密连接蛋白不仅是维持上皮屏障功能的重要蛋白,更是多种病毒感染细胞的受体与辅助分子。为了明确紧密连接主要组成蛋白occludin和claudin-1在鸭坦布苏病毒(Duck Tembusu virus, DTMUV)入侵细胞中的作用,本试验利用荧光定量PCR方法对病毒感染的非洲绿猴肾(vero)细胞两基因mRNA的表达量进行动态检测。结果表明,occludin和claudin-1的表达量与病毒含量具有相关性,感染后48 h内随着病毒含量的增加,occludin和claudin-1的mRNA上调表达,当病毒拷贝数下降且细胞出现明显病变后,两基因变为下调表达。试验结果有助于明确occludin和claudin-1在介导DTMUV入侵细胞中的作用,为后续功能受体的筛选和抗病毒制剂的研发提供依据。Tight junction proteins are not only important for maintaining epithelial barrier function but also receptors and accessory molecules of many viruses infect cells. In order to define the roles of occludin and claudin-1, major components of tight junction proteins, in Duck Tembusu virus(DTMUV) invasion cells, the dynamic changes of mRNA expression of the two genes in vero cells were detected by fluorescence quantitative PCR(qPCR). The results indicated that the mRNA expression level of occludin and claudin-1 were correlated with the virus content. With the increase of virus content within 48 hours after infection, the mRNA expression of occludin and claudin-1 was up-regulated, and then downregulated with the cytopathic effect(CPE) emergence and virus copies decreasing. These findings would help clarify the role of occludin and claudin-1 in mediating DTMUV invading cells and provide references for identification of functional receptors and development of antiviral reagents for DTMUV.
