摘 要:为探究天农麻鸡腹脂沉积候选基因表达水平和腹脂率的关系,本研究屠宰126日龄天农麻鸡200只,剥离腹部脂肪并称重,按腹脂率高低排序,随机选取不同腹脂率的32只鸡,提取腹脂组织的总RNA并利用qRT-PCR定量检测10个脂肪沉积候选基因的表达量,分析各基因表达量与腹脂率的相关性,并统计候选基因表达量在高低腹脂率天农麻鸡腹脂中的差异性。结果表明,LPL、PLIN、FTO和PPARγ表达量随腹脂率增加而逐渐高表达,其中LPL、PLIN和FTO表达量与腹脂率呈显著正相关且表达量在极端组间差异显著,可能是正向调节天农麻鸡腹部脂肪沉积的重要候选基因。该结果为研究天农麻鸡腹脂沉积的遗传调控机制和筛选分子标记提供了科学依据。The aim of this study is to explore the relationship between the expression levels of candidate genes for abdominal fat deposition in Tiannong partridge chicken and the abdominal fat percentage(AFP). A total of 200 Tiannong partridge chicken were slaughtered at 126-day-old. Then abdominal fat tissue was weighed and collected. Sorted by AFP, 32 chickens with different abdominal fat ratio were randomly selected for further analyses. Total RNA of abdominal fat tissue from the selected samples were extracted and quantitatively detected the expression levels of 10 fat deposition candidate genes by qRTPCR. Afterwards, the correlation between the expression level of each gene and AFP, as well as the differences of candidate gene expression levels between the high and low AFP chicken were analyzed. The results showed that the expression levels of LPL, PLIN, FTO and PPARγ gradually increased along with the increase of abdominal fat deposition in Tiannong partridge chicken. Among them, the expression levels of LPL, PLIN and FTO were significantly positively correlated with AFP of Tiannong partridge chicken(P<0.05), and their expression levels were significantly different between the high and low AFP groups(P<0.05). The mentioned genes might be important candidates for positive regulation of abdominal fat deposition in Tiannong partridge chicken. This result provided a scientific basis for the studies on genetic regulation mechanism and molecular markers of abdominal fat deposition in Tiannong partridge chicken.
