摘 要:为了明确喜马拉雅旱獭骨骼肌对高原低氧环境适应的组织学特点,本试验采用H.E.染色、免疫组织化学和电镜技术,对喜马拉雅旱獭骨骼肌的组织学指标进行观测与分析,并与SD大鼠进行比较。结果显示:喜马拉雅旱獭的骨骼肌肌纤维直径小于SD大鼠,且差异极显著(P<0.01),喜马拉雅旱獭的骨骼肌肌纤维表面积密度大于SD大鼠,且差异显著(P<0.05);喜马拉雅旱獭的骨骼肌血管内皮生长因子(VEGF)和微血管密度(MVD)均高于SD大鼠,且差异极显著(P<0.01);对于骨骼肌线粒体平均截面积、平均体积,喜马拉雅旱獭均小于SD大鼠,且差异极显著(P<0.01),对于体积密度、面数密度,喜马拉雅旱獭均高于SD大鼠,且差异极显著(P<0.01)。结果表明:喜马拉雅旱獭骨骼肌对高原低氧环境表现出良好的适应性,主要表现为骨骼肌的肌纤维直径细,肌纤维表面积密度大,VEGF大量表达,MVD大,线粒体体积密度和面数密度高。In order to determine the histological characteristics of the skeletal muscle of Himalayan marmot adapted to the high altitude hypoxia environment, H.E. staining, immunohistochemistry and electron microscopy techniques were used to observe and analyze the histological indexes of the skeletal muscle of Himalayan marmots, in comparison with SD rats. The results showed that the diameter of skeletal muscle fibers of Himalayan marmots was smaller than that of SD rats and the difference was extremely significant(P<0.01),and the surface density of skeletal muscle fibers of Himalayan marmots was greater than that of SD rats and the difference was significant(P<0.05). Vascular endothelial growth factor(VEGF) and microvessel density(MVD) of Himalayan marmots were higher than those of SD rats, and the difference was extremely significant(P<0.01). The average cross-sectional area and volume of skeletal muscle mitochondria of Himalayan marmots were smaller than those of SD rats, and the difference was extremely significant(P<0.01). The bulk density and surface number density of Himalayan marmots were higher than those of SD rats and the difference was extremely significant(P<0.01). Thus, Himalayan marmot skeletal muscle demonstrates good adaptability to the plateau hypoxia environment, mainly manifested as thin muscle fiber diameter, large muscle fiber surface area density, abundant expression of VEGF,large MVD,and high mitochondrial volume density and surface number density.
