摘 要:为探究鸭肠炎病毒(DEV)和A型产气荚膜梭菌(CpA)共感染的致病机制,本研究以DEV贵州株(DEV GZ)和CpA标准株(CVCC2030)分别及共感染37日龄健康雏鸭,采用病原分离鉴定和PCR检测确定成功感染后,采用液相色谱-质谱联用(LC-MS)技术检测DEV感染组、CpA感染组以及二者共感染组感染90 h时鸭十二指肠的代谢组学,筛选并分析潜在的差异代谢物及相关信号通路。结果显示:与对照组相比,DEV感染组感染90 h时鸭十二指肠有2’-脱氧核苷和苯丙酮酸等52种差异代谢物;CpA感染组有青霉素和3-羟基丁酸等29种差异代谢物;DEV和CpA共感染组有花生四烯酸和吲哚-3-乳酸等50种差异代谢物。DEV感染组鸭十二指肠在正负离子模式下,差异代谢物主要富集的代谢通路均为22条,且主要为氨基酸的生物合成代谢通路;CpA感染组鸭十二指肠在正离子模式下,差异代谢物主要富集的代谢通路为25条,且主要为类固醇激素的生物合成代谢通路;共感染组鸭十二指肠在正负离子模式下,差异代谢物主要富集的代谢通路分别为15条和14条,其中与免疫相关的代谢通路有色氨酸代谢和半胱氨酸-蛋氨酸代谢等。这些差异代谢物提示花生四烯酸和吲哚-3-乳酸可能成为DEV和CpA共感染鸭十二指肠的潜在生物标记物,本研究为阐明DEV与CpA共感染的致病机制提供了科学依据。To investigate the pathogenesis of co-infection of duck enteritis virus(DEV) and Clostridium perfringens type A(CpA). In this study, 37-day-old ducklings were infected with duck enteritis virus Guizhou strain(DEV GZ) and CpA(CVCC2030).And Liquid chromatography-mass spectrometry(LC-MS) was used to detect duodenal metabolomics of ducks in DEV infection group, CpA infection group and coinfection group at 90 hours time point. Potential differential metabolites and related signaling pathways were screened and analyzed.The results showed that compared with normal control group,there were 52 kinds of total differential metabolites in duodenum in DEV infected group at 90 hours post infection,including 2’-deoxynucleoside and phenylpyruvate.CpA infected group had 29 kinds differential metabolites induding penicillin and 3-hydroxybutyric acid.In DEV and CpA coinfection groups,there were 50 kinds differential metabolites induding arachidonic acid and indole-3-lactic acid.In the duodenum of DEV infected group,22 metabolic pathways were mainly enriched in differential metabolites under positive and negative ion mode,which were mainly amino acid biosynthesis.Under the positive ion mode,the duodenum of CpA infected ducks was mainly enriched in 25 metabolic pathways,which were mainly the biosynthesis of steroid hormones.In the coinfection group,there were 15 and 14 metabolic pathways enriched in differential metabolites in duodenum under positive and negative ion mode,respectively,among which immune-related metabolic pathways were tryptophan metabolism and cysteine-methionine metabolism.These differential metabolites suggested that arachidonic acid and indole-3-lactic acid might be potential biomarkers for DEV and CpA coinfection in duodenum of ducks,providing a scientific basis for elucidating the pathogenic mechanism of DEV and CpA coinfection.
