摘  要:目的探讨白及多糖(BSP)对烟曲霉菌诱导的小鼠巨噬细胞炎症反应的调节作用。方法通过CCK-8细胞毒性实验检测0、1、2、4、8、16、32、64、128 g/L的BSP溶液对巨噬细胞活力的影响;烟曲霉菌灭活菌丝感染RAW264.7细胞后给予16 g/L的BSP溶液进行处理,应用RT-PCR方法检测其炎症因子白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)和模式识别受体LOX-1 mRNA的表达;应用ELISA和Western blot方法检测炎症因子IL-1β、IL-6、TNF-α和模式识别受体LOX-1蛋白的表达。结果不超过32 g/L的BSP不影响RAW264.7细胞活性(F=0.08~2.09,P>0.05)。BSP可以显著降低烟曲霉菌刺激所引起的巨噬细胞模式识别受体LOX-1和下游炎症因子IL-1β、IL-6和TNF-α的mRNA表达(F=341.20~1112.33,P<0.001)和蛋白表达(F=74.20~641.34,P<0.001)。结论BSP通过下调巨噬细胞中模式识别受体LOX-1表达,抑制下游促炎因子表达,发挥对烟曲霉菌感染后的抗炎作用。Objective To investigate the regulatory effect of Bletilla striata polysaccharides(BSP)on the inflammatory response of mouse macrophages induced by Aspergillus fumigatus.Methods CCK-8 cytotoxicity assay was used to investigate the influence of BSP on macrophage viability at the concentrations of 0,1,2,4,8,16,32,64,and 128 g/L.RAW264.7 cells were infected by the inactivated hyphae of Aspergillus fumigatus and then treated with 16 g/L BSP solution;real-time PCR was used to measure the mRNA expression levels of the inflammatory cytokines interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)and the pattern recognition receptor LOX-1,and ELISA and Western blot were used to measure the protein expression levels of the inflammatory factors IL-1β,IL-6,and TNF-αand the pattern recognition receptor LOX-1.Results BSP with a concentration of≤32 g/L did not affect the viability of RAW264.7 cells(F=0.08-2.09,P>0.05).BSP significantly reduced the mRNA and protein expression levels of the macrophage pattern recognition receptor LOX-1 and the downstream inflammatory cytokines IL-1β,IL-6,and TNF-αinduced by Aspergillus fumigatus stimulation(mRNA expression:F=341.20-1112.33,P<0.001;protein expression:F=74.20-641.34,P<0.001).Conclusion BSP exerts an anti-inflammatory effect after Aspergillus fumigatus infection by downregulating the pattern recognition receptor LOX-1 in macrophages and inhibiting the expression of downstream proinflammatory factors.
