皮肤扁平苔藓中miR-27b与相关蛋白表达及意义

摘　　要：目的探讨皮肤扁平苔藓(CLP)中microRNA-27b(miR-27b)与相关蛋白的表达水平及意义。方法采用实时荧光定量PCR(RT-PCR)方法,检测40例CLP病人(CLP组)皮损组织及40例健康人(Con组)正常皮肤组织中miR-27b的表达水平。采用酶联免疫吸附测定(ELISA)法,检测两组皮肤组织中白细胞介素1β(IL-1β)和基质金属蛋白酶2(MMP-2)的表达水平。采用Spearman相关分析法分析CLP组皮损组织中miR-27b、IL-1β、MMP-2表达水平之间的相关性。结果与Con组相比,CLP组皮损组织中miR-27b表达水平明显下调(t=2.501,P<0.05),IL-1β和MMP-2表达水平明显上调(t=-2.383、-2.487,P<0.05)。Spearman相关性分析显示,CLP组皮损组织中miR-27b与IL-1β、MMP-2表达水平呈负相关(r=-0.361、-0.385,P<0.05),IL-1β与MMP-2表达水平呈正相关(r=0.332,P<0.05)。结论miR-27b及其相关蛋白可能参与了CLP的发病过程。Objective To investigate the expression levels and significance of microRNA-27b(miR-27b)and related proteins in cutaneous lichen planus(CLP).Methods Quantitative real-time PCR was used to measure the expression level of miR-27b in the skin lesions of 40 CLP patients(CLP group)and the normal skin tissue samples of 40 healthy controls(Con group).ELISA was used to measure the expression levels of interleukin 1β(IL-1β)and matrix metalloproteinase-2(MMP-2)in skin tissue of the two groups.A Spearman correlation analysis was used to analyze the correlation between the expression levels of miR-27b,IL-1β,and MMP-2.Results Compared with the Con group,the CLP group had a significant reduction in the expression level of miR-27b(t=2.501,P<0.05)and significant increases in the expression levels of IL-1βand MMP-2 in skin lesion(t=-2.383,-2.487;P<0.05).The Spearman correlation analysis showed that in the CLP group,miR-27b was negatively correlated with IL-1βand MMP-2(r=-0.361,-0.385;P<0.05),and IL-1βwas positively correlated with MMP-2(r=0.332,P<0.05).Conclusion This study shows that miR-27b and related proteins may be involved in the pathogenesis of CLP.
