摘  要:目的 分析C-X-C基序趋化因子配体(CXCL)9/10在头颈鳞状细胞癌(HNSCC)中的表达情况,并探讨其与HNSCC患者临床病理特征、人乳头瘤病毒16(HPV16)感染的相关性。方法 收集60例HNSCC组织标本和其中30例相应癌旁组织标本。采用实时荧光定量PCR检测HNSCC组织和癌旁组织中CXCL9/10 mRNA表达水平;采用免疫组化分析CXCL9/10在HNSCC组织和癌旁组织中的阳性表达情况;采用单管荧光定量PCR检测HNSCC组织中HPV16阳性率。分析CXCL9/10表达水平与HNSCC患者临床病理特征、HPV16感染情况的关系。结果 HNSCC组织中CXCL9、CXCL10 mRNA表达水平和阳性表达率均高于癌旁组织(均P<0.05)。CXCL9、CXCL10高表达患者中TNMⅢ~Ⅳ期、淋巴结转移及HPV16阳性的患者比例均高于低表达患者(均P<0.05)。结论 CXCL9/10在HNSCC组织中呈高表达,且CXCL9/10高表达的HNSCC患者的TNM分期更晚,可能存在淋巴结转移且感染HPV16的风险更高。Objective To analyze the expressions of C-X-C motif chemokine ligand(CXCL)9/10 in head and neck squamous cell carcinoma(HNSCC) and their correlation with clinicopathological features and human papillomavirus 16(HPV16) infection in HNSCC patients.Methods The HNSCC tissue specimens of 60 cases and paracancerous tissue specimens of 30 cases therein were collected.The expressions of CXCL9/10 mRNA in HNSCC and paracancerous tissues were detected by the real-time fluorescent quantitative PCR.The immunohistochemistry was used to analyze the positive expressions of CXCL9/10 in HNSCC and paracancerous tissues.The single-tube fluorescent quantitative PCR was performed to detect the positive rate of HPV16 in HNSCC tissues.The relation between CXCL9/10 expressions and clinicopathological features and HPV16 infection in patients with HNSCC was analyzed.Results The mRNA expressions and positive rates of CXCL9/10 in HNSCC tissues were higher than those in paracancerous tissues(all P<0.05).The patients with high-expressions of CXCL9/10 exhibited higher proportions of the patients in TNM Ⅲ-Ⅳ stage,with lymph node metastasis,and in HPV16 positive as compared with those with low-expressions(all P<0.05).Conclusion CXCL9/10 are highly expressed in HNSCC tissues,and the HNSCC patients with high CXCL9/10 expressions interpret an advanced TNM stage and have higher risk of lymph node metastasis presence and HPV16 infection.
