摘 要:以茖葱鳞片为外植体,研究不同培养基对愈伤组织诱导和植株再生的影响.结果表明:茖葱鳞片在MS培养基中诱导的愈伤组织好于White和WPM培养基,说明MS提供的营养物质更有利于茖葱愈伤组织生长.在MS培养基中加入6-BA与2,4-D或NAA不同用量组合都可以诱导茖葱鳞片形成愈伤组织.其中,MS+6-BA 1.5 mg/L+2,4-D 0.3 mg/L和MS+6-BA 1.5 mg/L+NAA 0.2 mg/L诱导的茖葱鳞片愈伤组织效果较好;6-BA与2,4-D或NAA组合诱导的茖葱愈伤组织都能够分化出不定芽,而6-BA与NAA组合诱导的不定芽数量较多;在MS+6-BA 1.5 mg/L+NAA 0.2 mg/L培养基中不定芽诱导率最高,达到97.66%,不定芽增殖数量(5.16个)较多;茖葱芽苗在1/2MS+NAA 0.5 mg/L+6-BA 0.1 mg/L中不定根发生率最高(平均99.12%)、单株根数最多(平均5.12条);以松针土为基质时,茖葱组培苗成活率最高(平均96.58%).Using Allium victorialis L.bulb as explants,the effects of different media on callus induction and plant regeneration were studied.The results show that the callus growth in MS medium is better than that in White and WPM medium.The results showed that the nutrients provided by MS were more beneficial to the growth of Allium victorialis callus.Combination of adding different dosages of 6-BA and 2,4-D or NAA in MS medium can induce to form callus.Among them,MS+6-BA 1.5 mg/L+2,4-D 0.3 mg/L and MS+6-BA 1.5 mg/L+NAA 0.2 mg/L have better effects on inducing callus of Allium victorialis bulb.The callus induced by 6-BA and 2,4-D or NAA can be differentiated into adventitious buds,but the number of adventitious buds induced by 6-BA and NAA was more.The induction rate of adventitious buds in MS+6-BA 1.5 mg/L+NAA 0.2 mg/L was the highest,reaching 97.66%,and the number of adventitious buds was 5.16.In 1/2 MS+NAA 0.5 mg/L+6-BA 0.1 mg/L,the adventitious root incidence rate(99.12%on average)and the number of roots per plant(5.12 on average)were the highest.When pine needle soil is used as substrate,the survival rate of tissue culture seedlings of Allium victorialis is the highest(96.58%on average).
