摘 要:实验室条件下利用东营盐碱土壤浸泡水培养螺旋藻(Spirulina platensis)的可行性研究。首先以东营盐碱土壤浸泡水加入氮源质量浓度为2.5g/L、磷源质量浓度为0.5g/L及其他Zarrouk培养基微量元素的培养基可以用于螺旋藻培养。光强为120μmol·m^(-2)·s^(-1)、全光照(24:0)、3%(v/v)CO_(2)为螺旋藻的最适培养条件,其15d培养生物量可达11~12g/L,固碳速率达到2.21g·L^(-1)·d^(-1);基于经济成本考量,直接用盐碱土壤浸泡水中仅添加适当的氮源和磷源和通入3%CO_(2)(v/v)的简易培养基下螺旋藻的生长情况:加入0.5g/L K_(2)HPO_(4)和2.5g/L NaNO_(3),螺旋藻蛋白质含量和多糖分别达到45%和16%,从而验证了利用盐碱地大水漫灌土工作业改造时的盐碱浸泡水培养螺旋藻来实现固碳并提升土壤有机质的技术是有效的。The feasibility of Spirulina platensis cultivation with extracted water from Dongying saline soil was experimental investigated.Firstly,the main factors such as the concentrations of nitrogen(NaNO_(3))and phosphorus(K_(2)HPO_(4)),light intensity,light-dark period and CO_(2) concentration in aerated gas on the growth of S.platensis were discussed separately by the Zarrouk medium from Dongying saline soil extracted water be used to instead of its solution.Results show Dongying saline soil water can guarantee Spirulina platensis cultivation.The biomass of S.platensis reached 11~12 g/L and the carbon fixation rate of 2.21 g·L^(-1)·d^(-1)after 15 days cultivation.while appropriate conditions of 2.5 g/L NaNO_(3),0.5 g/L K_(2)HPO_(4),120μmol·m^(-2)·s^(-1)and 24 h irradiance,and 3%CO_(2)enriched gas aeration.Considering the additives cost,results demonstrated that simple medium prepared by only adding NaNO_(3)and K_(2)HPO_(4)into saline soil water was also supported the growth of S.platensis under 3%CO_(2)enriched gas aeration.The contents of both crude protein and polysaccharides in S.platensis cultivated in simple medium by only adding 2.5 g/L NaNO_(3),0.5 g/L K_(2)HPO_(4)for 8 days were got 45%and 16%respectively.The technology will be effective that cultivated S.platensis be used to fixed carbon and improved soil organic matter by the way of extracted water from Dongying saline soil with ecological rehabilitation project.
